摘要 目的对大连地区152例RhD阴性血孕妇的RHD基因进行分型,并分析不同RHD基因型的Rh C、c、E和e表型分布特征。方法用PCR-SSP法对孕妇的RHD基因进行分型;分别应用单克隆抗-C、抗-c、抗-E和抗-e鉴定孕妇的Rh表型。结果共发现105例RHD全缺失型,以ccee表型(85.7%)为主;19例RHD 1227A纯合型,以Ccee表型(78.9%)为主;14例RHD-CE(2-9)-D2型,均为Ccee表型;6例弱D15型,均有c和E抗原;4例RHD 1227A杂合型,均有C和e抗原;4例用此PCR-SSP法无法判断分型结果。结论大连地区RhD阴性孕妇的RHD基因型呈现多样性,其中大多数为RHD全缺失型,其次为RHD 1227A纯合型和RHD-CE(2-9)-D2型。 Objective To determine RHD genotypes and characteristics of RhC/c/E/e phenotypes of 152 RhD-negative pregnant women in Dalian.Methods RHD gene was genotyped by PCR-SSP method.RhC/c/E/e phenotypes of the pregnant women were detected with monoclonal Anti-C,Anti-c,Anti-E and Anti-e antibodies.Results 105 cases of RHD deletion were identified with ccee as the predominant phenotype(85.7%).19 cases were determined as RHD 1227 A homozygotes with predominant Ccee phenotype(78.9%).All of 14 cases of RHD-CE(2-9)-D2were identified with Ccee phenotype.6 cases of weak D type 15 with cE and 4 cases of RHD 1227 A heterozygotes with Ce were identified.In addition,4 cases could not be genotyped by the PCR-SSP method.Conclusion Diversity of RHD genotypes is found in the pregnant women in Dalian.Most pregnant women are RHD deletion genotype,followed by RHD 1227 A homozygotes and RHD-CE(2-9)-D2.
