三种血细胞分离机采集单个核细胞后献血者的血常规变化

Changes of blood routine indonors after apheresis of peripheral blood mononuclear cells by using 3 kinds of blood cell separators

目的探索健康志愿者使用3种不同血细胞分离机采集单个核细胞后血常规的变化。方法分别使用3种不同品牌的血细胞分离机Fenwal Amicus、Spectra optia、MCS+9000采集了志愿者20人次的外周血单个核细胞,单采前、采后即刻(0 h)、1 h、48 h分别检测献血者的血常规指标。结果 3种机型的采集时间分别为:(2.58±0.19)h、(2.83±0.37)h、(3.33±0.37)h。MCS+处理血量最少,小于5 000 mL,其余两者均在7 000 mL以上。志愿者的WBC、Hb及Plt计数均在单采后有所下降,单采前和采后0 h、1 h、48 h存在统计学的差异(F=6.304、33.737、25.859,P均<0.05)。3种机型间相比,WBC、Hb及Plt单采前后的变化无显著差异(P>0.05)。结论单采PBMC完成后即刻,白细胞、血红蛋白和血小板计数会有所下降,但是1 h后迅速回升,证明使用这3种血小板分离机采集健康志愿者的PMBC对其血常规指标没有较大影响。

Objective To explore the changes of blood routine parameters in healthy volunteers after apheresis of peripheral blood mononuclear cells(PBMCs)by using 3 kinds of blood cell separators.Methods 20 apheresis of PBMC were performed with three different blood cell separators:Fenwal Amicus,Spectra optia and MCS+9000.Hb,Plts and WBCs of blood donors were detected before apheresis and at 0 h,1 h and 48 h after apheresis respectively.Results The duration of apheresis for three separators were(2.58±0.19)h,(2.83±0.37)h and(3.33±0.37)h.The total processed blood volume was less than 5 000 mL for MCS+9000 and above 7 000 mL for Fenwal Amicus and Spectra optia.WBC,Hb and Plt counts of donors decreased at 0 h after apheresis,and there were significant differences of WBC,Hb and Plt counts among before apheresis,at 0 h,1 h and 48 h after apheresis(F=6.304、33.737、25.859,P<0.05).However,there was no significant diference in terms of changes of WBC,Hb and Plt counts during the three separators.Conclusion WBC,Hb and Plt count of donors decreased at different levels at 0 h after PBMC apheresis,but returned quickly at post-apheresis 1 h,which proved that there is no obvious influence on blood routine parametersof donors who experienced PBMC apheresis by using the three different blood cell separators.
