摘要
目的探讨寨卡病毒(Zika Virus,ZIKV)与不同血型红细胞的粘附作用。方法选取A型、B型、O型及AB型全血各5 U,离心后去除白膜层与血浆,红细胞经生理盐水洗涤后重悬于等体积的同型血浆,添加一定比例的寨卡病毒,置37℃孵育箱孵育。3 d后,分别留取各血液成分血样,其中红细胞以生理盐水洗涤后重悬。抽提全血、红细胞以及血浆中的核酸并定量检测病毒载量。结果所有血型的红细胞和血浆中均能够检测到载量>10~8 copies/mL的寨卡病毒RNA;且O型红细胞中的RNA拷贝数高于血浆,差异具有显著性(P<0.05);而AB型红细胞中的RNA拷贝数则显著低于血浆(P<0.05)。结论寨卡病毒对红细胞具有一定的粘附作用,且该作用在不同血型的红细胞间存在差异。
Objective To investigate the adherence of Zika Virus(ZIKV)to different blood types of red blood cells.Methods Five units for each blood type of A,B,O and AB whole blood were selected.After centrifugation,buffy coat and plasma were removed,and red blood cells(RBCs)were suspended in plasma of the same type with the same volume.ZIKV suspension was added to the above samples in a certain proportion,and then mixed and incubated in an incubator at 37℃.Three days later,the whole blood samples and the upper plasma by centrifugation were collected detected for ZIKV RNA,and the RBCs were washed and resuspended with normal saline followed by viral load detection.Results ZIKV(>10~8 copies/mL)was detected in all RBCs and plasma samples.ZIKV RNA quantification were significantly higher than those in plasma(P<0.05)in type O RBCs and lower than those in plasma(P<0.05)in type AB RBCs.Conclusion ZIKV adheres to erythrocyte,and the effect of adhesion is vary between RBCs of different blood types.
