加味人参乌梅汤及其拆方对腹泻大鼠结肠黏膜病理形态学的影响 被引量:1

Effects of Modified Renshen Wumei Decoction and its Decomposed Formulas on Pathomorphology of Colonic Mucosa in Diarrhea Model Rats

摘要 目的:观察加味人参乌梅汤及其拆方对腹泻大鼠结肠黏膜病理形态学变化的影响。方法:SD大鼠96只,雌雄各半,随机分为空白组、模型组、自然恢复组、妈咪爱组、全方组、去甘味药组、去酸味药组、去辛味药组,每组12只。空白组常规喂养,其余各组采用苦寒中药泻下、游泳力竭法复合制作腹泻模型,连续造模14天。造模成功后,空白组和模型组用纯净水20ml·kg^(-1),自然恢复组常规喂养7天自然恢复,其余各组分别予:西药组予妈咪爱ig(0.7g·Kg^(-1)),加味人参乌梅汤全方ig(2.32 g·Kg^(-1)),去甘味药方ig(2.32g·Kg^(-1)),去酸味药方ig(2.32g·Kg^(-1)),人参乌梅汤去辛味药方ig(2.32g·Kg^(-1))。均1次/d,连续7天,取各组大鼠结肠组织,在光学显微镜下进行组织病理形态学观察。结果:模型组肠黏膜多处溃疡、坏死,黏膜下层有明显充血、水肿,灶样炎细胞浸润,各治疗组结肠黏膜炎性侵润均有改善,其中以加味人参乌梅汤去甘味药组效果最好。结论:加味人参乌梅汤对腹泻模型大鼠具有生津止泻作用,对结肠黏膜溃疡与破坏的腺体的改善可能是其重要机制之一。 Objective : To investigate the effect of modified renshen wumei decoction and its decomposed formulas on pathomorphology of colonic mucosa in diarrhea model rats. Methods : Ninety-six SD rats ( half male and female) were randomly divided into control group, model group, natural recovery group, positive control group (medilac-vita group), modified renshen wumei decoction group, removing sweet drugs group, removing sour drugs group, removing heat drugs group, 12 per group. The control group was fed conventionally. In other groups, the diarrhea models were established by multi-factors which includes the cold diarrhea TCM drugs and swimming exhaustive method. Continuous modeling for 14 days. After the modeling, the control group and model group were given normal saline ig ( 20 mL ·kg^-1) ; the natural recovery group was fed conventionally for 7 days, then naturally recovered; the rest of groups were respectively given (positive control group) Medilac-Vita ig(0. 7 g · Kg^-1) ,modified renshen wumei decocotion ig (2. 32g ·kg^-1), modified renshen wumei decocotion without sweet drugs ig (2.32 ·kg^-1), modified renshen wumei decocotion without sour drugs ig(2. 32g ·kg^-1) , modified renshen wumei decocotion without heat drugs ig(2. 32g ·kg^-1), 1 time daily, continuously for 7 days. To take the colon tissue of rats in each group and observe the histopathological morphology under the optical microscope. Results: Model group showed that multiple ulcers and necrosis occurred in the intestinal mucosa, and there were obvious congestion, edema, stove - like inflammatory cell infiltration in the submucosa. Meanwhile, inflammatory invasions of colonic mucosa in each treatment group were all improved. Among them, the group of modified renshen wumei decocotion without sweet drugs showed the best effect. Conclusion:Flavored ginseng mume decoction have the fluid antidiarrheal effect on diarrhea model rats and the treatment of colonic mucosa, while the effect of group of modified renshen wumei decocotion without sweet drugs is the best effect. The destruction on the gland may be one of the important mechanisms. Its pharmacological mechanism remains to further study.
