摘要
对纤维素酶辅助提取紫苏叶挥发油的工艺进行优化。采用单因素试验考察酶解时间、酶解pH、酶解温度及酶用量对挥发油得率的影响,得到最佳工艺条件为酶解时间45min、pH 4.6、温度55℃、酶用量0.0202g/10g(紫苏叶),在最佳条件下挥发油的得率为0.409%。再利用Design-Experts 8.0进行响应面实验设计,研究了各变量交互作用及其对紫苏挥发油得率的影响。结果表明:pH和酶用量属于显著性影响因子,对pH和酶用量的进一步分析表明在实验空间中存在响应面,其极大值点为pH 4.84、酶用量0.0213g/10g(紫苏叶),在极大值点可获得最高紫苏油得率为0.4896%,较单因素的实验结果提高了19.7%。
The cellulase assisted extraction of volatile oil irom Perilla frutescens leaves is optimized. Single factor experiment is used to investigate the effects of enzyme hydrolysis time, pH, cellulase dosage and temperature on the extraction efficiency of volatile oil, and then the variable interaction and its effects on extraction efficiency of volatile oil are researched by Design-Experts 8.0 and response surface methodology. The results show that pH and cellulose dosage are significant influence factors. With the further analysis, there's a response surface in the experiment space and the site of maximum value is pH 4.84 and cellulase dosage of 0. 0213 g/10 g (Perilla frutescens leaves). The maximum product rate is 0.4896% under the site of maximum value and it is 19.7% higher than the result of single factor experiment.
