摘要
目的建立HPLC-UV法同时测定猪胆粉中牛磺猪去氧胆酸和甘氨猪去氧胆酸的含有量。方法猪胆粉甲醇提取液采用Kromasil C_（18）色谱柱（4.6 mm×250 mm,5μm）分析;流动相乙腈-0.03 mol/L磷酸二氢钠,梯度洗脱;体积流量1.0 m L/min;柱温25℃;检测波长200 nm。结果牛磺猪去氧胆酸和甘氨猪去氧胆酸分别在0.087 7~4.384 9μg（r=1.000 0）、0.257 8~8.594 6μg（r=1.000 0）范围内呈良好的线性关系,平均加样回收率在95.0%~105.0%之间,RSD均小于3.0%。结论该方法简单、可靠,适用于猪胆粉的质量控制。
AIM To establish an HPLC method for determining taurohyodeoxycholic acid and glycohyodeoxycholic acid in Pulvis Fellis Suis. METHODS The analysis of Pulvis Fellis Suis methanolic extract was performed on Kromasil C_（18）column（ 4. 6 mm × 250 mm,5 μm）,mobile phase was acetonitrile-0. 03 mol / L sodium dihydrogen phosphate with gradient elution,flow rate was 1. 0 m L / min,column temperature was maintained at 25 ℃,and detection wavelength was set at 200 nm. RESULTS Taurohyodeoxycholic acid and glycohyodeoxycholic acid had good linear relationships in the ranges of 0. 087 7- 4. 384 9 μg（ r = 1. 000 0） and 0. 257 8- 8. 594 6 μg（ r =1. 000 0）,respectively. The average recoveries were 95. 0%- 105. 0%. The RSDs were less than 3. 0%. CONCLUSION This method is simple and reliable,which is suitable for the quality control of Pulvis Fellis Suis.
