摘要
为探究引起江苏某渔场泥鳅发病死亡的病原,本研究对其病变组织进行细菌分离,获得一株优势生长细菌;通过对该菌株的形态观察、生理生化特性检测、16S r RNA基因测序比对及系统发育树聚类,结果表明该分离株为霍乱弧菌(V.cholerae),将其命名为BH-1;回归试验证实该菌为致病菌,能够导致健康泥鳅死亡;药敏试验结果表明该分离菌株对氨曲南、强力霉素、庆大霉素等29种药物高度敏感,对头孢唑啉、青霉素G、乙酰螺旋霉素等6种药物中度敏感,对苯唑西林、万古霉素、林可霉素等8种药物已产生耐药性。本研究确定霍乱弧菌BH-1为引起该渔场泥鳅大量死亡的病原,本研究结果为该病防治提供实验依据。
In order to identify the pathogen which caused a serious mortality of raised loach (Misgurnus anguillicaudatus) occurred in some farms in Jiangsu province during August 2014, a predominant bacteria was isolated from ascites and ulcers of these diseased fishes. And the isolate was a high pathogenicity to the loach, which caused the similar clinical symptoms in artificially infected loach as the naturally infected cases. Finally, the bacteria isolate was identified as Vibrio cholerae by Gram-stained, phenotypic characteristics, artificial infection and 16S rRNA gene analysis. Furthermore, the drug susceptibility test with Kindy-Bauer antiotic testing showed that among the test 43 antibiotics, the bacteria isolate was sensitive to 29 antibiotics, moderate sensitive to 6 antibiotics and resistant to 8 antibiotics. These results provided basis for efficient control of the disease in loach.
