摘要
为系统评价云南越桔属资源的遗传多样性及亲缘关系,以零南省农业科学院园艺作物研究所蓝莓试验基地保存的13份云南越桔属资源叶片为材料,采用CTAB提取方法,对越桔基因组DNA进行了提取,并对获得的DNA进行了琼脂糖凝胶电泳检测及含量测定分析。结果表明:11份越桔叶片中提取出基因组DNA,且DNA的纯度和完整性都较好,质量高,D260nm/D280nm的比值均在1.8~2.0;2份野生越桔材料未获得谱带。
For system evaluation of genetic diversity and genetic relationship of blueberry resource, the genomic DNA of leaves of 13 blueberry (Vaccinium spp. )species were isolated by CTAB method,which from Institute of Horticultural Crops,Yunnan Academy of Agricultural Sciences,and the purity and quality of each DNA ob- tained was evaluated by agarose gel electrophoresis and UV scanning. The results showed that the genomic DNA extracted from leaves of 11 blueberry(Vaccinium spp. )species by this method was pure,integral, the val- ue of D260 nm/D280 nm was 1.80 to 2.0. Two wild species fail to get genomic DNA band.
