摘要
目的探讨羟喜树碱(HCPT)对宫颈癌HeLa细胞作用的机制。方法 利用western blot、GFP—LC3 shRNA转染、Hoechst染色等对经HCPT处理后的HeLa细胞进行自噬和凋亡水平的观察。结果western blot、荧光显微镜的结果均提示,HCPT能够诱导HeLa细胞中自噬相关基Beclin-1、p62以及LC3的表达增加,从而激活细胞自噬发生;除细胞自噬外,HCPT也可以造成细胞的凋亡,且凋亡出现在自噬触发之后。结论HCPT能够激活宫颈癌HeLa细胞发生自噬,进而诱导细胞凋亡达到抗肿瘤目的。
Objective To explore the antitumor mechanism of Hydroxycamptothecin ( HCPT ) on cervical cancer. Methods Western blotting and Hoechest staining Methods were used to detect the level of apoptosis and autophagy after HCPT being used on Hela which is cervical cancer cell line. Results After using HCPT, the expression of the regulators ofBeclin-1, p62 and microtubule-associated protein 1 light chain 3 ( LC3 ) were upregulated and then the autophagy of Hela cells were triggered. In the meanwhile, the autophagy induced cell apoptosis. Conclusion HCPT can induce Hela cells apoptosis by stength the cells autophagy.
