虾夷扇贝组织结构分析

Tissue structure analysis of patinopecten yessoensis

把虾夷扇贝作为实验材料,掌握虾夷扇贝血涂片的制作方法,并运用HE染色组织切片的方法对虾夷扇贝的组织结构进行观察以及分析。用5mL注射器吸取2mL肝素钠,抽取1mL左右虾夷扇贝心脏血液,从载玻片一端滴一滴血液,左手水平拿着玻片,右手用另一个载玻片水平轻推血液,使其展平,在阴凉处风干,同样方法制取五个血涂片。留一个做空白对照,取两个血涂片用瑞氏染液染色,其他两个吉姆萨染液染色,放于阴凉处晾干留作观察。取新鲜扇贝组织切成1.0cm×1.0cm×0.3cm的组织块,经过固定、脱水与透明、浸蜡、包埋等过程后把组织块切成4-6μm的石蜡切片。再经过苏木素染色盐酸酒精褪色,后用伊红染液复染等过程后用中性树胶完成封片,待干后进行显微镜的观察以及拍照。最后通过观察与人体以及鱼类相关的组织结构进行分析对比。

The experimental materials of patinopecten yessoensis were the experimental materials,master the method of making blood smear for patinopecten yessoensis,the tissue structures of patinopecten yessoensis were observed and analyzed by hematoxylin-eosin staining and tissue section.With 5mL syringe absorbs the heparin sodium 2mL,then extraction of grass carp 1mL of blood,on one end of the slide drip a drop of blood,the lift hand flat,right hand with another level of slide force will promote blood,air dry in the shade,make five blood smear in the same way.After drying,take one blood smear used as blank control,take two blood smear dyeing with Switzerland,another two blood smear with Jim dyeing liquid dye,air dry in the shade,wash,observed.Take the fresh tissue of patinopecten yessoensis tissue was cut into 1.0＊1.0＊0.3cm of the Scallop in Shell,after immobilization,dehydration and transparent,leaching wax,embedding process,the tissue block cut into 4~6μm paraffin sections.Then after hematoxylin staining hydrochloric acid alcohol to fade,with eosin stain re dyeing process with neutral gum complete mounting and dried for microscope observation and photography.Finally,the structure of the human body and fish was analyzed and compared with the observation and comparison.
