摘要
[目的]探讨丹蛭降糖胶囊干预2型糖尿病大鼠血管并发症的机制。[方法]采用高糖高脂饲料联合STZ注射制备2型糖尿病大鼠模型,观察丹蛭降糖胶囊干预8周后模型大鼠主动脉形态学改变、主动脉组织AGEs、RAGE和血清VCAM-1、ICAM-1含量的变化。[结果]高糖高脂饲料联合STZ注射后模型大鼠腹主动脉出现显著病理学改变,腹主动脉AGEs、RAGE含量显著升高(P<0.01),血清VCAM-1、ICAM-1含量显著升高(P<0.01);而丹蛭降糖胶囊各剂量组大鼠的腹主动脉病理学改变得到不同程度改善,腹主动脉AGEs、RAGE含量显著降低(P<0.05或P<0.01),血清VCAM-1、ICAM-1含量显著降低(P<0.05或P<0.01)。[结论]丹蛭降糖胶囊能够有效防治2型糖尿病模型大鼠主动脉病理损伤,其干预机制与调节AGEs-RAGE通路密切相关。
[Objective ]To explore the mechanism of Dartzhi Jiangtang Capsule (DJC) for intervening vascular complications in type 2 diabetic rats. [Methods]The type 2 diabetic rats model was made by feeding high-sugar and high-lipid fodder and injecting streptozotocin (STZ). Eight weeks after DJC intervention, aortic morphological changes, variation of AGEs, RAGE in aorta tissue and the level of serum VCAM-1 ,ICAM-1 were observed. [Results]Significant pathological changes could be seen in abdominal aortic of rats model after feeding high-sugar and high-lipid fodder and injecting STZ. There is significant increase showed in abdomi- nal aortic AGEs, RAGE (P〈0.01) and in the levels of serumVCAM-I,ICAM-1 (P〈0.01). While improved pathological changes of abdominal aortic to varying degrees could be seen in DJC groups of each dose. There is significant decrease showed in abdominal aortic AGEs, RAGE (P〈0.05 or P〈0.01 ) and in the levels of serumVCAM-1 ,ICAM-1 (P〈0.05 or P〈0.O1 ). [Conclusions] DJC can prevent aortic pathological damage of type 2 diabetic model rats effectively. The intervening mechanism of DJC is strongly associated with the regulation of AGEs-RAGE pathway.
