丹参酮ⅡA对ox-LDL诱导血管内皮细胞自噬效应蛋白Beclin1表达的影响 被引量:11

摘要 目的氧化型低密度脂蛋白(oxidized low-densitylipoprotein,ox-LDL)可诱导血管内皮细胞发生自噬,造成血管内皮损伤,从而导致动脉粥样硬化的发生。该研究拟探讨丹参酮ⅡA对ox-LDL诱导的血管内皮细胞自噬效应蛋白Beclin1表达的影响。方法 (1)运用不同浓度ox-LDL处理人脐静脉血管内皮细胞HUVECs,MDC染色、流式细胞仪检测ox-LDL对HUVECs自噬水平的影响,采用免疫印迹技术检测自噬标记蛋白Beclin1表达;(2)ox-LDL处理后,运用不同浓度丹参酮ⅡA干预,MDC染色、流式细胞仪HUVECs自噬水平变化,免疫印迹检测Beclin1蛋白水平变化。结果 ox-LDL能诱导HUVECs发生自噬,丹参酮ⅡA能抑制ox-LDL诱导的HUVECs自噬效应蛋白Beclin1表达。结论丹参酮ⅡA通过抑制ox-LDL诱导的HUVECs自噬,从而对ox-LDL所致的HUVECs损伤起保护作用。 Objective Oxidized low- densitylipoprotein( ox- LDL) can induce the autophagy of vascular endothelial cell and cause damage to it,which results in atherosclerosis. This study is to explore the effect of Tanshinone ⅡA on the expression of autophagy protein Beclin1 of vascular endothelial cell induced by ox- LDL. Methods We used different densities of ox- LDL to deal with the human umbilical vein vascular endothelial cells( HUVECs),then used the MDC staining,flow cytometry( FLM) to detect the effect of ox- LDL on autophagy level of HUVECs and used western blot to detect the expression of autophagy protein Beclin1. After the induction of ox- LDL,different densities of Tanshinone ⅡA were used to intervene,then the MDC staining,flow cytometry( FLM) were used to detect the change in autophagy level of HUVECs and western blot was used to detect the expression of autophagy protein Beclin1. Results Ox- LDL could induce the autophagy of HUVECs and Tanshinone ⅡA could inhibit the expression of autophagy protein Beclin1 of HUVECs induced by ox- LDL. Conclusion Tanshinone ⅡA has a protective effect on the damage to HUVECs caused by ox- LDL by inhibiting the autophagy of HUVECs induced by ox- LDL.
