摘要
目的探讨人促黄体激素释放激素(luteinizing hormone releasing hormone,LHRH)受体在喉癌组织中的表达及其在喉癌HEp-2细胞膜表面与LHRH-PE40的亲和力。方法取外科手术收集的50份喉鳞癌及20份癌旁组织标本,采用Western blot法检测LHRH受体的表达,免疫荧光分析法检测LHRH受体的亚细胞定位,ELISA法检测HEp-2细胞膜表面LHRH受体与LHRH-PE40的亲和力。结果 LHRH受体在喉癌组织中的表达量明显高于癌旁组织(P<0.001);LHRH受体在喉癌组织中定位于细胞膜和细胞质,而在癌旁组织中定位于整个腺体周边;LHRH受体在HEp-2细胞膜表面与LHRH-PE40的结合显示出线性和饱和性,且可被LHRH直接特异性抑制。结论 LHRH受体在喉癌组织中的过度表达及其在喉癌细胞表面与LHRH-PE40的高亲和力表明,其可能是LHRH-PE40的一个潜在的治疗靶点。
Objective To investigate the expression of luteinizing hormone releasing hormone(LHRH) receptor in larynocarcinoma tissue as well as its affinity to LHRH-Pseudomonas aeruginosa exotoxin 40(PE40) on the surface of HEp-2 cells. Methods Fifty specimens of larynocarcinoma tissue and 20 specimens of adjacent noncancerous tissue collected by surgery were tested for expression of LHRH receptor by Western blot, for subcellular location of LHRH receptor by FA, and for affinity of LHRH receptor to LHRH-PE40 on the surface of HEp-2 cells by ELISA. Results The expression level of LHRH receptor in laryngocarcinoma tissue was significantly higher than that in adjacent noncancerous tissue(P〈0. 001). LHRH receptor was located in the cytomembrane and cytoplasma of laryngocarcinoma tissue, while in the surrounding glands of adjacent noncancerous tissue. The affinity of LHRH receptor to LHRH-PE40 showed linearity and saturability, which was inhibited directly and specifically by LHRH. Conclusion The overexpression of LHRH receptor in laryngocarcinoma tissue and the high affinity of LHRH to LHRH-PE40 indicated that the receptor might be a potential target of LHRH-PE40.
