牛樟芝ITS序列分析及液体发酵菌丝体总三萜含量测定 被引量:9

ITS Sequence Analysis of Antrodia cinnamomea and Total Triterpenoid Detection of Liquid Fermentation Mycelium

摘要 为筛选富萜牛樟芝(Antrodia cinnamomea)菌株,对收集的3株牛樟芝菌株(AC620,AC1404,AC001)进行ITS序列分析,通过香草醛-冰醋酸法对牛樟芝液体发酵菌丝体总三萜含量进行分析。ITS序列分析结果表明菌株AC1404和AC001的ITS序列完全相同,长度均为593 bp,AC 620菌株的长度为581 bp。供试菌株AC001、AC1404和AC620菌丝体中的三萜含量分别为34.30、18.47和16.68 mg·g-1(DW),菌株AC001与AC1404和AC620间菌丝三萜含量差异极显著(P<0.01)。 In order to screen the rich triterpenoid Antrodia sp.,3 collected Antrodia cinnamomea strains( AC620 AC1404,AC001) were analyzed with ITS sequence. The triterpenoid contents of Antrodia cinnamomea in liquid fermentation mycelium were tested by the method of vanillin- glacial acetic acid. It showed that the ITS sequences were the same in the strains AC 1404 and AC 001,the length was 593 bp,and was 581 bp of strain AC 620. The triterpenoid contents of tested strains were 34. 30,18. 47 and 16. 68 mg · g- 1( DW) in strain AC001,AC1404 and AC620,respectively. There was significant difference( P 0. 01) between AC1404,AC620 and AC001.
