鹅PRLR基因克隆及在生殖相关组织中的表达规律 被引量:6

Cloning of PRLR Gene and Its mRNA Expression Regulation of Tissue Correlated with Reproduction in Goose

摘要 本文采用克隆、测序技术获得浙东白鹅PRLR基因c DNA序列,利用实时荧光定量PCR技术检测鹅产蛋期和就巢期PRLR基因在生殖相关组织中的表达规律,为揭示PRLR基因在生殖过程中的功能提供资料。结果表明,在获得的浙东白鹅PRLR基因c DNA序列中,其中编码区2496 bp可编码831个氨基酸,与其他鹅PRLR序列相比发生了T667C、G738A、C757T、G1047A突变,其中T667C引起氨基酸R-C的变化,C757T处引起氨基酸W-R的变化。在产蛋期和就巢期时,垂体中PRLR基因mRNA表达量均显著性高于下丘脑、卵巢和输卵管,其他组织间表达量没有显著性差异。同一组织在产蛋期和就巢期之间存在显著差异,部分组织达极显著差异。本文试验结果为继续开展鹅的就巢性方面的研究提供了一些数据。 In this study,the c DNA of PRLR gene in Zhedong white goose by cloning and sequencing technique was obtained. The mRNA expression rule of PRLR gene in tissue which was correlated with reproduction in laying period and broodiness period by Real-Time PCR was analyzed to provide information for the function of PRLR gene in reproductive process. The result showed that the PRLR gene c DNA of Zhedong white goose was obtained,which contained 2496 bp with coding region of encoding 831 amino acids. Compared with sequence of other goose PRLR gene,there were four mutations: T667 C,G738A,C757 T and G1047 A,of which T667 C and C757 T resulted in the amino acid variations of R-C and W-R,respectively. In laying period and broodiness period,the PRLR gene mRNA expression level of hypophysis was significantly higher than that of hypothalamus,ovary and oviduct. The mRNA expression was not significantly different among hypothalamus,ovary and oviduct. The mRNA expression in the same tissues was significantly different between laying period and broodiness period. The mRNA expressions of some tissues were very significantly different. The result would provide the reference for ovary research of goose.
