摘要
研究猪瘟兔化弱毒苗在免疫猪体内的动态分布,在注射猪瘟兔化弱毒苗后第1、7、14、21、28、35、63天采集免疫猪的脾、肾、肺、腹股沟淋巴结、扁桃体及血清,应用免疫组化、RT-PCR、猪瘟抗原、抗体检测试剂盒等方法,检测分析猪瘟病毒在猪体内的动态分布、存留时间及抗体水平。结果表明:应用免疫组化方法于免疫后第7~14天在脾脏、第7~21天在腹股沟淋巴结、第14~28天在肾脏组织分别检测到猪瘟病毒。在免疫后第7天检测到猪瘟病毒抗体,随后抗体水平持续上升至免疫后第63天;病毒抗原的分布与RT-PCR检测结果吻合。结论:猪瘟兔化弱毒苗在猪体内主要分布器官为脾、肾和腹股沟淋巴结,免疫后第7天脾和腹股沟淋巴结最早检测到猪瘟病毒,猪瘟病毒在肾停留时间可至免疫后第28天。实验过程中未在扁桃体、肺、外周血检测到猪瘟病毒。
In order to study the dynamic distribution and retention times after immuning pigs with classical swine fever virus( CSFV) lapinised vaccine,spleen,kidney,lung,lymph nodes,tonsil and serum of immune pig were collected in the 1,7,14,21,28,35,63 days post immunization( dpi),and the dynamic distribution,retention time of CSFV in pigs as well as the antibody levels induced by CSFV were detected with the immuno-histochemistry( IHC) method,RT-PCR,serum antigen and antibody analysis. CSFV antibody showed positive in the7 dpi,antibody levels continued to rising until to 63 dpi. CSFV antigen was detected positive in the spleen by IHC after 7dpi to 14 dpi,also in the groin lymph nodes from 7 dpi to 21 dpi,and in kidney during 14 dpi to 28 dpi. Further more,the distribution of virus antigen agree to the RT-PCR results. CSFV could not be detected in the tonsil,lung and peripheral blood throughout the experiment. The CSFV mainly distributes in the organs such as spleen,kidney and inguinal lymph nodes,CSFV was first detected in spleen and inguinal lymph node at 7 dpi,and may be stay in the kidney until 28 dpi.
