摘要
OBP18是蜜蜂气味结合蛋白质(odorant binding proteins,OBPs)家族中的成员,参与气味分子的识别,对嗅觉起着重要的作用。为研究蜜蜂OBP18基因功能及与抗螨性状的相关性,本研究以中华蜜蜂头部组织c DNA为模板,PCR扩增OBP18基因完整CDS序列,利用生物信息学方法分析其CDS序列及其编码的氨基酸序列,用RT-PCR方法检测OBP18基因在不同抗螨性状的中华蜜蜂头部组织中的相对表达量。结果表明:OBP18基因CDS全长为399 bp,编码132个氨基酸的分泌性蛋白,无跨膜螺旋,是比较保守的基因;RT-PCR检测在螨虫胁迫下的蜂螨抗性中华蜜蜂头部组织中OBP18基因表达量显著高于未受胁迫的抗性群体,及受到同等蜂螨胁迫的敏感性群体,显示中华蜜蜂OBP18基因的表达变化与蜂螨感染过程相关,提示OBP18基因在中华蜜蜂的抗螨行为中起着重要作用。
OBP18 was one member of family of odorant binding proteins( OBPs),which was involved in the identification of odor molecules and plays an important role in olfaction. In order to survey the function of the OBP18 gene and its relationship with Varroa destructor resistance,the complete coding sequence( CDS) of the OBP18 gene was obtained by PCR amplification using the c DNA from the head tissue of Chinese honeybee,then the CDS of the OBP18 gene and its amino acid sequence were analyzed by bioinformatics method,and then the RTPCR assay was used to determine the relative expression level of the OBP18 gene in the head of Chinese honeybees with different Varroa destructor resistance. The results showed that the CDS of the OBP18 gene was 399 bp in size,encoding for a protein of 132 amino acids. The encoded protein was a secretary protein without transmembrane domain. The OBP18 gene was a relatively conserved gene. It was found that the OBP18 gene had higher expression levels in the head of resistant Chinese honeybee colony when being challenged by Varroa destructor resistant to parasitism than that in the unchallenged resistant colony and sensitive colony,which indicated that OBP18 might be involved in the progress of mite resistance of Apis cerana cerana.
