猴头菌中三种脂肪酸含量HPLC-ELSD测定方法的建立 被引量:1

摘要 建立猴头菌(Hericium erinaceus)子实体中棕榈油酸(palmitoleic acid)、异油酸(vaccenic acid)、软脂酸(palmitic acid)的高效液相色谱一蒸发光散射检测(high performance liquid chromatography—evaporative light—scattering detector,HPLC—ELSD)方法。采用Grace Prevail Organic Acid5μm(250mm×4.6mm)色谱柱[流动相为乙腈.甲醇(80:20),柱温30℃,流速1.0mL/min]和ESA Chromachem蒸发光散射检测器[雾化温度(TNcb)35℃,蒸发温度(TEvap)50℃,衰减值(Attenuation)为3,载气为N2,操作气压为21psi],获得棕榈油酸、异油酸、软脂酸标准品的线性回归方程分别为Y:0.4982x+10.1120(r=0.9998)、Y=0.8270X+9.7457(r=0.9998)、Y=1.1316x+10.3790(r=0.9997);平均回收率分别为99.89%、100.68%和101.00%,RSD分别为1.26%、2.04%和1.72%。该方法可同时测定猴头菌中棕榈油酸、异油酸、软脂酸3种脂肪酸类化合物,且结果准确、重现性好、操作简便。 A High Performance Liquid Chromatography-Evaporative Light-Scattering Detector (HPLC- ELSD) system that was simple, accurate and exhibited good reproducibility, was used to determine palmitoleic, vaccenic and palmitic acid levels in petroleum ether extracts of Hericium erinaceus fruit bodies. The system employed a Grace Prevail Organic Acid 5 μm analytical column (250 mm × 4.6 mm, column temperature 30 ℃) and acetonitrile : methanol (80:20, flow rate 1. 0 mL/min) as the mobile phase. Operating conditions for the ESA Chromachem ELSD system were as follows: nebulization temperature (TNeb) 35 ℃, evaporation temperature(TEvap)50 ℃, attenuation 3, carrier gas N2, carrier gas pressure 21 psi. Linear concentration ranges and average recovery rates for palmitoleic, vaccenic and palmitic acids were 0.05 - 0.50μg/mL ( r = 0. 9998), 0.03 - 0.30μg/mL ( r = 0. 9998) and 0.02-0.20μg/mL ( r = 0.9997) respectively, and 99.89% (RSD = 1.26%), 100.68% (RSD = 2.04%) and 101.00% (RSD = 1.72%), respectively.
