摘要
类风湿关节炎(Rheumatoid arthritis,RA)是一种抗原驱动、T细胞介导及与遗传因素相关的慢性、炎症性自身免疫性疾病。白细胞介素(IL)-1β和肿瘤坏死因子α(TNF-α)等致炎性细胞因子在其发生发展过程中起重要作用,主要通过核因子NF-κB途径、MAPK途径、PI3K途径和JAK-STAT途径进行信号转导。而JAK/STAT信号通路是近年发现的重要的信号转导通路,参与调控人体多种器官和组织的发育、生长和分化。研究证实,在RA起病和关节破坏过程中,JAK/STAT信号通路发挥着重要的调控作用。JAK/STAT信号通路通过多种途径参与炎性反应,与RA关系密切。因此,了解JAK/STAT信号通路与RA之间的关系,能为开发新的抗RA药物提供新的理论依据。
Objective : Rheumatoid arthritis is an chronic inflammatory and autoimmune disease, driven by antigen, mediated by T cells and related to genetic factors: IL- 1 and TNF-a and other proinflammatory cytokines play an important role in the occurrence and development process, mainly through nuclear factor kappa B pathway ( NF- κB ) , MAPK pathway, PI3K pathway and JAK-STAT pathway of signal transduction. JAK-STAT signal pathway is a recently identified important signal transduction pathway. The development, growth and differentiation are involved in the regulation of various body organs and tissues. The study confirmed that, in the process of RA onset and joint destruction, JAK-STAT signaling pathway plays an important role in the regulation. JAK-STAT signal pathway is involved in the inflammatory resp.onse through a variety of ways, with tight relationship with RA. Therefore, understanding the regulation of the relationship between JAK-STAT signaling pathway and RA could provide new opportunities for the development of new anti RA drugs.
