摘要
文章对昆明小鼠特应性皮炎模型的建立进行了研究。目的:制备昆明小鼠的特应性皮炎模型。方法:模型组分别于第1、8、15天用2%DNCB(橄榄油配置)100μL涂于背部去毛区域,第18、19、20天用0.5%DNCB 10μL涂于模型右耳激发,对照组用橄榄油替代DNCB。观察各组小鼠皮损情况并进行动态分级,计算耳肿胀度、胸腺指数、脾指数,HE染色观察皮肤炎性改变。结果:模型组表皮红肿增厚,有脱屑和渗出。模型小鼠耳肿胀度、胸腺指数、脾指数明显高于对照组(P<0.05)。HE染色模型小鼠局部皮肤肥厚,炎细胞浸润等炎性改变明显,对照组未发现异常。结论:2%DNCB(橄榄油配置)可用于昆明小鼠特应性皮炎的制备。
This paper researches on the establishment of atopic dermatitis model on Kunming mice aiming to prepare Kunming mice model of atopic dermatitis. Methods: applying 100 ul 2% DNCB(olive oil configuration) on the model group back without hair area at day1, day8, and day15 respectively, on 18 th, 19 th, 20 th day using 10 ul 0.5%DNCB at right ear to excitation, the control group was treated with olive oil. Observing the lesions situations and making dynamic classifications, calculating ear swelling, thymus and spleen index, analyzing skin inflammatory changes through HE staining. Results: The skin of model mice is inflamed. The ear swelling, thymus index and spleen index of model mice significantly higher(P〈0.05) than control mice. HE staining shows local skin hypertrophy and other inflammatory reactions change significantly in model mice group, while the control group is normal.Conclusion: 2% DNCB(olive oil configuration) can be prepared atopic dermatitis on Kunming mice.
