摘要
目的:通过建立小鼠心肌缺血再灌注模型,分析白细胞介素(IL)-9在心肌缺血再灌注损伤(MIRI)中的作用。方法:用结扎小鼠冠状动脉左前降支(LAD)的方法制作IRI模型,并于再灌注前5 min颈静脉注射rIL-9,同时设立对照组。以Evan's blue和TTC双染色法来评价缺血区和梗死面积。应用电化学发光法检测血清中心肌肌钙蛋白T(cTnT)水平。通过超声心动图和血流动力学检测评价心功能。结果:再灌注后24h,rIL-9干预组和对照组小鼠心肌的梗死面积、血清cTnT水平、心功能均无显著差异。结论:IL-9并不会影响小鼠心肌缺血再灌注损伤的进程。
Objective:To investigate the effect of IL-9in myocardial ischemia-reperfusion injury(IRI)in mice.Method:The IRI model was performed by ligating the left anterior descending(LAD)of coronary artery in mice,with a jugular vein injection of rIL-9at 5min before reperfusion,and a control group was set up.Evan's blue and TTC double staining was performed to evaluate the AAR and infarct size.Serum cTnT levels were detected by electrochemical luminescence.The index of cardiac function was detected by echocardiography and hemodynamics.Result:In 24 hafter reperfusion,there was no significant difference in infarct size,serum TnT levels and cardiac function between the rIL-9injection group and control group.Conclusion:IL-9does not affect the process of myocardial ischemia reperfusion injury in mice.
