摘要 [目的]探讨不定芽的直接再生方式,建立高丛越橘组培快繁体系。[方法]以高丛越橘‘比洛克西’带腋芽茎段为外植体,在培养基中添加不同配比的ZT、6-BA、NAA和IBA,进行腋芽诱导、不定芽增殖和试管苗生根研究。[结果]消毒处理以0.1%Hg Cl24 min为宜,诱导率为68.3%;适宜不定芽继代增殖的培养基为WPM+ZT 2.0 mg/L,不定芽的增殖系数达到11.3;将试管苗转接至生根培养基WPM+IBA0.2 mg/L中,生根率为63.3%,平均每个苗形成4.8条根,根长9.4 mm。当试管苗长至5 cm时出瓶移栽,成活率可达80%以上。[结论]建立了高丛越橘组培快繁体系,为试管苗规模化生产奠定基础。 [Objective] To explore the direct regeneration way of adventitious bud,and establish tissue culture and rapid propagation of Vaccinium corymbosum. [Method]Using stems with auxiliary of Vaccinium corymbosum‘Biloxi'as explants,adventitious bud induction,proliferation,rooting of test-tube plantlets were observed on media supplied with various plant growth regulators and their combinations of ZT,6-BA,NAA and IBA. [Result]The results showed that fourminutes disinfection using 0. 1% mercuric chloride was the most proper for stem sterilization and induction rate of explants reached 68. 3%. The optimum medium for bud multiplication was WPM + ZT 2. 0 mg / L and the proliferation coefficient achieved to 11. 3. The rooting rate reached 63. 3%,and 4. 8 roots per plantlet and 9. 4 mm in root length were obtained when the plantlets in vitro were inoculated to rooting medium WPM + IBA0. 2 mg / L. The plantlets over 5 cm in length were transplanted and survival rate exceeded 80%. [Conclusion]The study establish tissue culture and rapid propagation of Vaccinium corymbosum,and lay the foundation for large-scale production of test-tube seedling.
