肉鸡鹦鹉热衣原体的分离鉴定及基因分型 被引量:1

Isolation and characterization of Chlamydia psittaci isolated from broilers

摘要 为了解肉鸡鹦鹉热衣原体感染流行情况并对其进行基因型分型,随机采集45日龄肉鸡的肺和脾,采用BGM(buffulo green monkey)细胞培养法进行病原分离培养,通过DNA微点阵列确定分离株的种属和基因型。结果显示,其中39.56%(36/91)的脾和45.39%(69/152)的肺样品的BGM细胞培养结果为阳性,鸡场感染率高达90%(9/10);通过种属DNA微点阵列成功鉴定确定72.46%(50/69)的肺分离株为鹦鹉热衣原体。基因型DNA微点阵列结果表明,鹦鹉热衣原体分离株的基因型为B型和D型。结论:所调查的鸡场鹦鹉热衣原体感染率高,流行的基因型主要为基因型B型和D型。本研究首次报道了采用细胞培养的方法从肉鸡组织器官中分离得到鹦鹉热衣原体,并用DNA微点阵列技术对其进行了基因分型,为肉鸡鹦鹉热衣原体病的流行病学调查提供了方法和手段。 The objective of this study was to investigate the genotypes of C.psittaci from broilers.C.psittaci was isolated from BGM cells of lungs and spleens,which were randomly collected from 45days-old broilers in slaughterhouse.The species and genotype of isolates were determined using DNA microarray.39.56% of the broiler spleens and 45.39% lungs were positive.Nine in ten(90%)of examined chicken farms were positive.The results of species DNA microarray showed that 72.46%of lung isolates were C.psittaci,and the genotype of lung isolates were belonged to genotype B and genotype D.This is the first time to report the isolation of C.psittaci from broiler lungs with BGM cells and identification of their species and genotypes by DNA microarray.It provides a method for epidemiological investigation of broiler chlamydiosis.
