摘要
为探究vasa基因在雄性大鼠性腺分化后生殖细胞发育中的作用,利用实时荧光定量PCR和免疫组织化学法对17.5日胎龄胎鼠和刚出生仔鼠的睾丸组织进行了vasa mRNA和VASA蛋白表达的检测。结果显示,17.5日胎龄胎鼠和刚出生仔鼠的睾丸组织均检测到vasa mRNA表达,且刚出生仔鼠的睾丸组织vasa mRNA表达相对较高。VASA蛋白只在刚出生仔鼠的睾丸组织表达,在17.5日胎龄胎鼠的睾丸组织未见表达。结果表明,vasa基因在生殖细胞发育过程中起重要作用,但作为标记物只能标记前精原细胞之后发育的各类生殖细胞。
In order to explore the role of vasa gene in the development of germ cells in male rats,in the present case,the expression of vasa mRNA and VASA protein was detected by Real time Q-PCR and immunohistochemistry method in 17.5days-old fetal rats and neonatal rats.In result,the expression of vasa mRNA was detected in the 17.5days-old fetal rats and neonatal rats,and the expression of vasa mRNA in testis of neonatal rats was high than the fetal rats.The expression of VASA protein was detected in the neonatal rats,But it was not detected in the fetal rats.In conclusion,vasa gene plays an important role in the development of germ cells.However,as a marker,it can only be used to develop all kinds of germ cells.
