基质金属蛋白酶-14与细胞外基质金属蛋白酶诱导因子在口腔鳞状细胞癌中的表达及临床意义 被引量:1

摘要 目的 探讨基质金属蛋白酶(MMP)-14和细胞外基质金属蛋白酶诱导因子(EMMPRIN)在口腔鳞状细胞癌(OSCC)中的临床意义,并分析二者的相关性。方法 选取2012年1月至2014年6月于我院口腔颌面外科诊治的69例OSCC患者作为OSCC组,同时选取43例口腔正常新鲜组织标本作为对照组。采用逆转录-聚合酶链反应(RT-PCR)技术检测组织中的MMP-14及EMMPRIN mRNA的表达含量。结果 MMP-14mRNA、EMMPRIN mRNA在OSCC组中的相对表达量明显高于其在对照组中的相对表达量,差异具有统计学意义(t=8.198、9.624,P0.05),在伴有淋巴结转移的患者中均明显升高(t=2.909、2.156,P〈0.05);且MMP-14、EMMPRIN mRNA在Ⅲ~Ⅳ期、中低分化的OSCC中表达均明显升高,差异具有统计学意义(t=3.376、2.667、3.027、2.399,P〈0.05);MMP-14与EMMPRIN mRNA表达含量间相关系数为r=0.821,二者呈正相关性(P〈0.01)。结论 OSCC组织中MMP-14和EMMPRIN的高表达状态均与肿瘤的病理分期、分化程度及淋巴结转移因素关系密切,两者的表达具有正相关性,提示MMP-14和EMMPRIN均参与了OSCC的浸润及转移过程,通过检测EMMPRIN及MMP-14mRNA表达含量可能有助于OSCC的病变程度及预后转归判断。 Objective To discuss the clinical relevance of MMP-14 and EMMPRIN expression in oral squamous cell carcinoma(OSCC). Methods Sixty-nine cases of patients with OSCC were chosen from January 2012- June 2014 in our hospital as the OSCC group, while 43 cases of patients with normal oral tissues were chosen as the control group. The expression levels of MMP-14 and EMMPRIN mRNA were detected by using RT-PCR. Results The expression of MMP-14 and EMMPRIN mRNA in OSCC group was significantly higher than that of the control group (t=8. 198, 9. 624, P〈0.01). As to the OSCC, there were no statistical differences in the expression of EMMPRIN and MMP-14 mRNA in age and gender (P〉0.05), while the expression increased significantly in lymph node metastasis, Ⅲ-Ⅳ stage and moderate or poor differentiation (P〈0.05) ; the correlation coefficient hetween the expression of mRNA MMP-14 and EMMPRIN was 0. 821, which indicated positive correlation (P〈 0.01). Conclusion The expression levels of MMP-14 and EMMPRIN mRNA in the OSCC have a close relationship with pathological stage, differentiation and lymph node metastasis, which shows the MMP-14 and EMMPRIN are involved in the invasion and metastasis of OSCC. It can be helpful to estimate the severity and the prognosis of the OSCC by detecting the expression of MMP-14 and EMMPRIN mRNA.
