摘要 [目的]建立延龄草愈伤组织诱导、增殖和分化再生体系。[方法]以延龄草块根、根尖为外植体,应用正交试验方法设计植物生长调节剂组合和质量浓度配比,研究植物生长调节剂对延龄草愈伤组织形成、增殖和分化的影响。[结果]1/2MS是最适合延龄草块根、根尖诱导愈伤的培养基,添加6-BA 1.2 mg/L和IAA 0.6 mg/L的1/2MS培养基最适合于愈伤组织的诱导,其中以根尖愈伤组织的诱导效果最好,在添加6-BA 0.5 mg/L、IAA 0.6 mg/L的培养基中,愈伤组织的分化率最高,附加6-BA 1.2 mg/L、IAA 1.0 mg/L的1/2MS培养基为生根适宜培养基。[结论]延龄草的块茎和根尖外植体在适宜的激素组合中均可通过愈伤组织途径分化出不定芽,继而得到正常生长、发育的再生植株。 [ Objective ] To establish the regeneration system of callus induction, proliferation and differentiation of TriUiumts chonoskii Maxim.. [ Method ] Tuber and root tip of Trilliumts chonoskii Maxim. were used as the explants, the composition and quality concentration ratio of plant growth regulators were designed by orthogonal experiment ,the effects of plant growth regulators on callus formation, proliferation and differentia- tion of Trilliumts chonoskii Maxim. were studied. [ Result] 1/2MS was a suitable culture medium of tuber and root tip of TriUiumts chonoskii Max- im.. 1/2MS Medium added with 6-BA 1.2 mg/L and IAA 0.6 mg/L was suitable for callus induction,the optimal examples were root tip. In me- dium added with 6-BA 0. 5 mg/L and IAA 0. 6 mg/L, the differentiation rate of callus was the highest. 1/2MS Medium added with 6-BA 1.2 mg /L, IAA 1.0 mg/L was suitable for rooting. [ Conclusion] Tubers and root tip explants of TriUiumts chonoskii Maxim. could be differentiated ad- ventitious buds by callus pathway in suitable hormone combination, and then get the normal growth and development of plant regeneration.
