Expression of NLRX1 in cochlear hair cells of C57BL/6 mice

摘要 目的探讨模式识别受体成员NLRX1在C57BL/6小鼠耳蜗毛细胞中的定位及其在小鼠生长发育、衰老过程中的动态表达。方法取健康新生4 d、1月龄、3月龄及9月龄C57BL/6小鼠耳蜗基底膜铺片,采用免疫荧光技术检测NLRX1在毛细胞内的定位及其随年龄的表达,采用RT-PCR、Western blotting法检测NLRX1在耳蜗毛细胞内的表达。结果新生4 d C57BL/6小鼠毛细胞不表达NLRX1;1月龄C57BL/6小鼠仅内毛细胞表达NLRX1,且主要位于纤毛;3月龄C57BL/6小鼠内、外毛细胞均表达NLRX1,纤毛部位强荧光信号;9月龄C57BL/6小鼠NLRX1表达明显下降,全部外毛细胞纤毛失表达,部分内毛细胞表达,伴有部分外毛细胞缺失。结论NLRX1随着C57BL/6小鼠发育逐步增加,随着年龄增加逐渐下降,推测NLRX1在毛细胞内的表达与听力的产生及维持可能具有一定关系。 Objective To explore the expression of nucleotide-binding domain and leucine-rich-repeat-containing family member X1( NLRX1) in the cochlear hair cells and its dynamic changes during the development and aging of C57 BL /6 mice. Methods Basilar membranes of C57 BL /6 mice in different age groups,4-day,1-month,3-month and 9-month,were obtained. The location and changes of NLRX1 expression were detected with immunofluorescence,Western blotting and real-time PCR. Results There was no expression of NLRX1 in neonatal mice on day 4 postnatally.The 1-month old mice only expressed NLRX1 in inner hair cells( IHCs),especially in the cilia. The 3-month old mice show ed NLRX1 expression in both IHCs and outer hair cells( OHCs). How ever,the 9-month old mice had markedly low er expression of NLRX1,which exhibited the entire lost expression in OHCs and partial lost expression in IHCs,with some OHCs missing. Conclusion The expression of NLRX1 in C57 BL /6 mice cochlear hair cells increases during development,but decreases in the process of aging. It is reasonable to speculate that NLRX1 expression in hair cells may be related to the formation and maintaining of hearing.
