摘要
研究在不同浓度下奶牛脐带间充质干细胞和奶牛乳腺上皮细胞共培养对共培养体系细胞增殖的影响。将P3代UC-MSCs与P3代BMECs按照浓度比例为1∶1、1∶2、1∶3、1∶4、1∶5、1∶10、1∶50、1∶100、1∶1000、2∶1等不同比例随机混合培养,同时设立UCMSCs与BMECs单纯培养组为对照组,并分别于0h,24h,48h,72h,96h,120h和144h时观察各组细胞的形态变化,用MTT法检测细胞增殖。将UC-MSCs和BMECs按照不同浓度比混合共培养后,细胞生长状态良好,无抑制作用;MTT检测增殖发现各组OD值在72h时均达到最大,其中1∶2浓度组OD值最大,细胞增殖最快。UC-MSCs和BMECs混合共培养,两种细胞之间无抑制作用,生长状态良好,同时能够促进共培养体系的细胞增殖。
The objective of the research is to study MSCs and BMECs co-culture on cell proliferation. In this the impact of different concentrations of UC- study, the P3 generation of UC-MSCs and P3 generation of BMECs with the concentration proportion of 1 :1, 1 : 2, 1 : 3, 1 : 4, 1 : 5, 1 : 10, 1 : 50, 1 : 100, 1 : 1000 and 2 : 1 etc. were cultivated randomly, and taking UC-MSCs and BMECs pure culture as the control group. The morphological changes of each group were observed at 0 h, 24 h, 48 h, 72 h, 96 h, 120 h and 144 h respectively by MTT cell proliferation assay. The results show the cell growth was in good condition, no inhibition occurred when the UC-MSCs and BMECs co-cultured with different concentrations according to the mixing ratio; MTT proliferation detection reveals OD values reached the peak at 72 h for each group, among which the group with a concentration of 1 : 2 was the highest and its cell proliferation was significantly other groups. Therefore, it is concluded that co-culturing UC-MSCs and BMECs can promote the proliferation system, in which UC-MSCs and BMECs do not inhibit each other and grow in a good condition.
