甘草次酸和EGF对HaCaT细胞miR-21/PDCD4表达的调控研究 被引量:1

摘要 目的探讨甘草次酸和表皮生长因子(EGF)对角质细胞(HaCaT)miR.21/PDCD4表达的调控作用。方法PcR法检测空白组、25ng/mlEGF组、25μm甘草次酸组、25ng/mlEGF联合25μm甘草次酸组对HacaT细胞miR-21、PDCD4mRNA表达的影响。结果25μm甘草次酸明显抑制HacaT细胞miR-21的表达水平,促进HacaT细胞PDcD4mRNA的表达;25μm甘草次酸抑制25ng/mlEGF对HacaT细胞miR.21的促表达作用,25μm甘草次酸抑制25ng/mlEGF对HacaT细胞PDcD4mRNA的下调作用。结论甘草次酸抑制HacaT细胞miR-21的表达、上调PDCD4mRNA的表达,同时可抑制EGF对HacaT细胞miR-21的促表达作用和对PDCD4mRNA的下调作用。 Objective To explore the effects of glycyrrhetinic acid and EGF on the expression of miR-21/PDCD4 in HaCaT cells. Methods PCR was used to detect in the blank group, 25ng/ml EGF group, 25 tx M glycyrrhetinic acid group, 25 ng/ml EGF and 25 μM glycyrrhetinic acid group on the effects of the expression of miR-21, PDCD4 mRNA in HaCaT cells. Results 25μM glycyrrhetinic acid significantly inhibited the expression level of miR-21 on HaCaT cell and promoted the expression of PDCD4 mRNA in HaCaT cell; 25 μ M glycyrrhetinic acid significantly inhibited the effect of 25ng/ml EGF on stimulating expression of miR-21 in HaCaT cell, and inhibiting the expression of PDCD4 mRNA in HaCaT cells. Conclusions Glycyrrhetinic acid inhibits the expression of miR-21 and stimulates the expression of PDCD4mRNA in HaCaT cells, and glycyrrhetinic acid inhibits the effect of EGF on stimulating the expression of miR-21 and down-regulates the expression of PDCD4 mRNA in HaCaT cells.
