人血浆中细胞外组蛋白在矽肺发病中的变化 被引量:4

The study on the change of extracellular histones in human plasma during the pathogenesis of silicosis

摘要 目的观察矽肺患者血浆中细胞外组蛋白水平,探讨细胞外组蛋白在矽肺病肺纤维化发生中的作用。方法选取矽肺患者62例作为矽肺组,其中I期23例,Ⅱ期25例,Ⅲ期14例;选取有2年以上职业性矽尘接触史但未诊断为矽肺的劳动者60例作为矽尘接触组;选取无职业性粉尘接触史的健康劳动者65例作为健康对照组。应用酶联免疫吸附方法测量血浆中细胞外组蛋白(H4)和转化生长因子.B(TGF.p)的浓度。结果与健康对照组[(0.82±0.67)μg/m1]比较,矽尘接触组[(4.14±2.85)g/m1]和矽肺组[(9.50±5.04)μg/ml】血浆中细胞外组蛋白H4含量明显升高,差异有统计学意义(P〈0.01)。血浆中的细胞外组蛋白H4水平与矽肺期别有明显的相关性(r=0.8955,问题丹0.0388)。矽肺组血浆中转化生长因子(TGF-β)浓度明显高于矽肺接触组和对照组,差异有统计学意义(P〈0.05)。矽肺患者血浆中细胞外组蛋白H4水平与TGF-β有明显的相关性(r=0.5375,P〈0.01)。结论血浆中细胞外组蛋白水平在矽肺发病过程中明显升高,细胞外组蛋白可能在矽肺纤维化进程中发挥重要作用。 Objective To investigate the plasma level of extracellular histones in patients with silicosis, and to explore the role of extracellular histones in the pathogenesis of pulmonary fibrosis in silicosis. Methods Sixty-two patients with silicosis were enrolled as the silicosis group, consisting of 23 patients with stage I silicosis, 25 with stage II silicosis, and 14 with stage III silicosis; sixty workers who had a history of occupational exposure to silica dust for more than 2 years and had not been diagnosed with silicosis were enrolled as the silica dust exposure group; sixty-five healthy workers without a history of occupational exposure to dust were enrolled as healthy controls. Enzyme-linked immunosorbent assay was applied to measure the plasma levels of plasma extracellular histone (H4) and transforming growth factor-β (TGF-β). Results Compared with healthy controls [(0.82±0.67) μg/ml], the silica dust exposure group [(4.14±2.85) μg/ml] and silicosis group [(9.50±5.04) μg/ml] had significant increases in plasma level of H4 (P〈0.01). The plasma level of H4 was significantly correlated with the stage of silicosis (r=0.8955, P =0.0388). The silicosis group had a significantly higher plasma level of TGF-β than the silica dust exposure group and healthy controls (P 〈0.05). In the patients with silicosis, the plasma level of H4 was significantly correlated with that of TGF-β (r=0.5375, P〈0.01).Conchtsion The plasma level of extraeellular histones increases significantly in the pathogenesis of silicosis, and extracellular histones may play an important role in the progression of fibrosis in silicosis.
