摘要
建立了快速检测发酵液中夫西地酸含量的高效液相色谱分析方法,采用Angilent Extend-C18色谱柱,用磷酸盐缓冲液—乙腈(30:70)作流动相,流速为1.2 m L·min-1,检测波长为214 nm,柱温为30℃。夫西地酸浓度在10μg^2 000μg·m L-1范围内,夫西地酸峰面积与质量浓度线性良好,相关系数为0.999 1;按标准加入法做回收率,平均回收率达到99.56%;精密度RSD为1.13%。该方法具有分析时间短、操作简便、结果准确,重现性好等特点,特别适用于大量的菌种筛选,提炼、纯化过程的在线监测。
HPLC( High performance liquid chromatography) method to determine the content of the fusidic acid in fermentation broth was developed. The HPLC was carried out with Agilent Extend- C18 column. The mobile phase was phosphate buffer- acetonitrile( 30∶70) with a flow rate of 1.2 m L·min^- 1. The detection wavelength was set at 214 nm and the column temperature was 30 ℃. The results show that the concentration of fusidic acid ranges in 10- 2 000 μg·m L- 1,the linear relationship between peak area and mass concentration is good, the correlation coefficient is 0.9991. According to standard addition method, the average recovery is 99.56% and the precision is 1.13%. The method is rapid in analysis,convenient in operation, accurate in result and good in reproducibility etc., and is suitable for a large number of strain screening and the online monitoring of refinery and purification.
