甲苯胺蓝-羧甲基壳聚糖-碳纳米管复合光敏剂的合成及体外灭菌性能 被引量:3

Synthesis of new photosensitizer-toluidine blue-carboxymethyl chitosan-carbon nanotube and its effects on inactivating periodontal pathogens in vitro

摘要 目的研究复合光敏剂甲苯胺蓝-羧甲基壳聚糖-碳纳米管(toluidine blue-carboxmethyl chitosancarbon nanotubes,TB-cm CS-CNTs)的特性及其对牙周致病菌的灭菌效果。方法选择门诊12例慢性牙周炎的患者,取其龈上菌斑进行体外培养,分别用TB和TB-cm CS-CNTs作为光敏剂对其进行光动力灭菌实验,光敏剂浓度均为1 mg/m L,光强度和光剂量分别为212 m W/cm2、12 J/cm2,分析比较TB和TBcm CS-CNTs的灭菌效果;检测TB-cm CS-CNTs的可见光吸收光谱并观察其在水中的稳定性。结果 TBcm CS-CNTs在可见光范围内的最强吸收峰位于610 nm处;在水中分散2周后仍然能保持较好的分散性,未出现明显沉淀;TB和TB-cm CS-CNTs的灭菌效率分别为(74.8±0.8)%和(81.4±1.0)%,差异具有统计学意义(P<0.05)。TB-cm CS-CNTs的暗毒性较低。结论 TB-cm CS-CNTs在水中具有较好的分散性和稳定性,相对于TB具有更好的灭菌效果。 Objective To evaluate the characteristics of new composite photosensitizer toluid- ine blue-carboxymethyl chitosan-carbon nanotubes (TB-CmCS-CNTs) and its effects on inactvi- ating periodontal pathogens in vitro. Methods TB-cmCS-CNTs visible absorption spectra was tested and its characterization in water was also observed. Twelve patients with untreated chronic periodontitis were selected, pthotodynamic therapy (PDT) was performed on with the concen- tration of photosensitizer (TB and TB-cmCS-CNTs) 1 mg/mL, light intensity was 212 mW/ cm2, and light dose was 12 J/cm2. Results The strongest absorption peak of TB-cmCS-CNTs in visible was located at 610 nm. It was still maintaining solubility well and no precipitation was found after soaking in water for two weeks. The efficacy of disinfection of TB-cmCS-CNTs and TB were ( 81.4± 1.0) % and (74.8 ± 0.8 ) %, respectively, and there was significam difference between the two groups (P 〈 0.05 ). Toxicity of TB-cmCS-CNTs was lower. Conclusion TB-cmCS-CNTs has better sterilization effect and more dispersibility and stability in water than TB.
