摘要
目的研究淋巴细胞相关微小核糖核酸(miRNAs)在肺炎支原体肺炎(MPP)患儿的表达差异。方法采用miRNA芯片技术筛选MMP患儿12例(A组)和非MPP患儿12例(C组)血液淋巴细胞中差异表达的miRNAs,RT-PCR方法验证芯片检测结果和检测参与淋巴细胞调节相关的特异性miRNAs表达情况。结果 A组初步建立MPP患儿血液淋巴细胞中miRNAs异常表达谱,筛选出105个上调表达的miRNAs和133个下调表达的miRNAs。与C组相比,A组30个miRNAs上调大于5倍,10个miRNAs上调大于10倍,133个miRNAs下调小于0.5倍(P<0.05)。A组miRNA-181a、miRNA-152-3p、miRNA-126-3p、miRNA-29c-3p和miRNA-20-3p表达水平均高于C组(P<0.05)。结论 miRNAs在MPP患儿血液淋巴细胞中存在差异表达,可能通过调节淋巴细胞功能参与免疫反应。
Objective To study the differential expressions of lymphocyte-related microRNAs(miRNAs)in children with mycoplasma pneumoniae pneumonia(MPP).Methods The differential expressions of miRNAs in lymphocytes of children with MPP(group A,12 cases)and non-MPP children(group C,12 cases)were screened by miRNA chip technique.The results were verified by RT-PCR chip and the expressions of lymphocyte-related miRNAs were detected in two groups.Results Abnormal expression spectrum of miRNAs in group A was initially built,in which 105 miRNAs were upregulated and 133 miRNAs were downregulated.Compared with group C,the upregulation of 30 miRNAs in group A was more than 5 times and that of 10 miRNAs was more than 10 times,but the downregulation of 133 miRNAs was less than 0.5 times(P〈0.05).Moreover,the expressions of miRNA-181 a,miRNA-152-3p,miRNA-126-3p,miRNA-29c-3p and miRNA-20-3p were higher in group A than those in group C(P〈0.05).Conclusion The differential expressions of miRNAs in blood lymphocytes of MPP children exist,which participate in the immune reactions by regulating the function of lymphocytes.
