摘要
目的探讨5-氨基乙酰丙酸-光动力学疗法(ALA-PDT)对体外培养的人皮肤鳞状细胞癌A431细胞增殖及其细胞周期的影响。方法以体外培养的人皮肤鳞状细胞癌A43l细胞株为研究对象,一组为对照组(不处理),另一组为ALAPDT组(0.4 mg/ml的ALA避光孵育3 h后行激光照射),银染细胞后镜下观察,并计算核仁组成区的相对面积;采用流式细胞术分析光动力学治疗后各细胞周期中细胞分布比例,并计算增殖指数。结果光动力学治疗后A43l细胞株细胞内核仁组成区的相对面积显著降低。流式细胞仪检测结果表明光动力学治疗组S期峰低于对照组,S期细胞比例显著降低为(9.2±2.1)%。对照组和光动力学治疗组的增殖指数分别为(47.0±3.4)%和(31.1±3.1)%,两者比较差异有统计学意义(P<0.05)。结论 5-氨基乙酰丙酸-光动力学疗法能够抑制A43l细胞株的过度增殖,改变其细胞周期分布。
Objective To investigate the effect of ALA-PDT on the proliferation and cell cycle of A431 cells. Methods A431 cells were cultured and divided into two groups. The cultured cells were treated with ALA-PDT in one group( ALA-PDT group),and not treated in the other group( control group). The relative surface of argyrophilic protein in nucleolar organizer regions( Ag NORs) was calculated after argyrophilic staining. Flow cytometry was applied to analyze the cell cycle and proliferation index( PI). Results The relative surface of Ag NORs in A431 cells was reduced markedly after ALA-PDT treatment. The cell percentage in S stage was decreased to( 9. 2 ± 2. 1) % in ALA-PDT group,significantly lower than in control group. PI in ALA-PDT group was less than that in control group[( 47. 0 ± 3. 4) % vs( 31. 1 ± 3. 1) %,P〈0. 05]. Conclusion ALA-PDT can inhibit the proliferation of A431 cells and change cell cycle of A431 cells.
