摘要 目的研究大孔树脂分离纯化黄刺玫果中总黄酮的工艺条件及参数。方法采用静态吸附解吸法考察8种大孔树脂对黄刺玫果中总黄酮的吸附和解吸性能,筛选树脂型号;以总黄酮的含量为指标考察各因素对D101大孔树脂纯化黄刺玫果中总黄酮的影响。结果选用D101型大孔吸附树脂,最佳工艺条件为:上样液料液比为1∶12,最大上样量为2.6 g生药/g树脂,用3BV的70%乙醇进行洗脱,解吸率达95%,总黄酮回收率为72%。经上述工艺纯化后黄刺玫果提取物中总黄酮含量为24.87%。结论该方法简单可行,分离效果好,可为工业生产提供参考。 Objective To optimize the process conditions of separation and purification of total flavonoids from Rosa xanthina lindl with macroreticular adsorption resin. Methods Static adsorption and desorption performance of eight kinds of macroporous adsorption resins for total fiavonoids from Rosa xanthina lindl were investigated to screen the optimal resin. The factors of D101 macroporous resin were investigated to purify the total flavonoids of Rosa xanthina lind, with the content of total flavonoids as the index. Results D101 macroporous adsorption resin was chosen as the optimal resin, and its optimized technology conditions were as following:the concentration of sample sotufion 1: 12, the largest sample weight of 2.6 g Rosa xanthina lindl/g resin, the volume of 70% ethanol(3BV)as the eluant. The desorption rate reached 95%, and the recovery rate of total flavonoids from Rosa xanthina lindl was 72%. After purified with DIO1 macroporous resin, the purity of total flavonoids was up to 24.87%. Conclusion This procedure is good and simple, and could he used to separate and purify the total flavonoids from Rosa xanthina lindl.
