摘要
目的研究载脂蛋白E(Apo E)与肿瘤发生之间的关系。方法建立小鼠肿瘤生长模型,观察野生型(WT)和Apo E基因敲除(Apo E KO)小鼠肿瘤生长情况,检测肿瘤小鼠脾脏中髓系抑制性细胞(MDSC)比例及与MDSC聚集相关因子的mRNA表达水平。结果 Apo E KO组肿瘤生长较WT组慢,脾脏内MDSC比例低于WT组,MDSC中白介素-6、转化生长因子-β和CC趋化因子受体2表达水平均低于WT组(P<0.05,P<0.01)。结论抑制Apo E表达可以阻碍MDSC的产生和聚集,进而抑制肿瘤生长。
Objective To investigate the relationship between apolipoprotein E (ApoE) and tumorigenesis. Methods The tumorous growth models of mice were obtained, and the conditions of tumor growth in rats with wild type (WT) and ApoE gene knock-out ( ApoE KO) were observed. The proportion of myeloid-derived suppressor cells (MDSC) in spleens and the mRNA expression level related MDSC accumulation were detected. Results In ApoE KO group, the tumor growth was slower, and the percentage of MDSC in the spleen, expression levels of interleukin-6 (IL- 6 ), transforming growth factor-β (TGF-β) and CC-chemokine receptor2 (CCR2) in MDSC were lower than those in WT group (P 〈 0. 05, P 〈 0. 01). Conclusion The tumor growth and accumulation of MDSC can be suppressed by inhibi- ting ApoE expression and tumor growth.
