2010年乌鲁木齐市腹泻婴幼儿轮状病毒感染基因特征哨点分析 被引量:2

Genetic sentinel characteristics of rotavirus infection among infants and children with diarrhea in 2010 in Urumqi

摘要 目的分析2010年乌鲁木齐市腹泻婴幼儿轮状病毒(rotavirus,RV)感染的流行情况及基因型特征。方法收集2010年1—12月乌鲁木齐市5岁以下门诊和住院腹泻患儿粪便样本1 001份,采用胶体金法和ELISA法检测RV,并随机选取约30%RV阳性样本进行G/P基因型分型。结果 1 001份样本中检出RV阳性样本336份,检出率为33.6%;随机选取100份阳性样本进行PCR分型,结果显示G1和G3型为主要流行G基因型,均占34.0%,其次为G9型(15.0%)和G2型(2.0%);G型混合感染占15.0%,以G1+G3为主(46.7%,7/15);P型常见的是P[8](67.0%),其次为P[6](8.0%)和P[4](3.0%);P型混合感染占21.0%,主要为P[8]+P[6](81.0%,17/21);P未能分型1份;主要的G/P基因型组合为G1P[8]和G3P[8],均占28.0%,其次是G9P[6](8.0%)、G9P[8](6.0%)和G2P[4](2.0%);其中2份新生儿RV阳性样本基因型均为G9P[6]型。结论 2010年乌鲁木齐市腹泻婴幼儿RV主要流行基因型为G1P[8]和G3P[8],应注意P[6]基因型可能引起的流行并需持续监测。 Objective To analyze the epidemiology and sentinel genotype characteristics of rotavirus infection among infants and children with diarrhea in 2010 in Urumqi. Methods A total of 1 001 stool samples from ≤5-year aged infants and children with diarrhea in January to October, 2010 in Urumqi were detected rotavirus by colloidal gold assay and ELISA method, and 300/oo of rotavirus positive samples were randomly selected for G/P genotyping. Results Rotavirus was detected in 336 out of 1 001 samples (33.6 %). Totally 100 positive samples were randomly chosen and genotyped by multiplex reverse transcription-PCR, which revealed G1 and G3 were the most prevalent types, accounting for 34.0% and 34.0%, followed by G9 (15.0~) and G2 (2. 0%). In G mixed infections (15.0%), G1 + G3 was the main types (46.7%, 7/15). In P type, P[8] was the main genotype (67.0%), followed by P[6] (8.0%) and P[4] (3.0%). P mixed infection accounted for 21.0%, in which P[8] +P[6] was the main type (81.0%, 17/21). One of P type was not typed. The major G/P combination was G1P[8] (28.0%) andG3P[8] (28.0%), followed byG9P[6] (8.0%), GgP[8] (6.0%) and G2P[4] (2.0%). Two neonatal RV strains were G9P[6] genotypes. Conclusion GIP[8] and G3P[8] are the main G/P combinations in infants and children in 2010 in Urumqi. P[6] genotype may cause prevalence and should be monitored continuously.
