摘要
目的研究马鞭草经皮透过液对B16黑色素瘤细胞的作用,在细胞水平上验证其具有美白活性。方法使用MTT法测定马鞭草经皮透过液对B16细胞增殖的影响;L-Dopa氧化法测定经皮透过液对B16黑色素瘤细胞内酪氨酸酶活性的影响;NaOH裂解法测定细胞内黑色素含量的影响;DAPI染色法检测细胞凋亡情况。结果马鞭草经皮透过液对B16黑色素瘤细胞的增殖、酪氨酸酶活性以及细胞内黑色素含量均有抑制作用,且其浓度越高抑制作用越强。结论马鞭草经皮透过液不仅影响B16黑色素瘤细胞的增殖,同时也影响细胞内酪氨酸酶的活性以及黑色素的合成,提示马鞭草具有对皮肤美白活性的功效,对开发成天然美白产品具有良好前景。
Objective To observe the effect of Herbal Verbenae percutaneous fluid on B16 melanoma cell and to verify its whitening activity at a cellular level. Methods The proliferation,tyrosinase activity and melanin biosynthesis of the B16 melanoma cells were measured by MTT cytotoxicity assay,L-dopa oxidation method and Na OH lysis assay,respectively. DAPI staining was used to detect the apoptosis of the B16 melanoma cells. Results Herbal Verbenae percutaneous liquid had dose-dependent inhibitory effect on the proliferation,tyrosine enzyme activity and melanin content of the B16 melanoma cells. Conclusion Herbal Verbenae percutaneous fluid not only affects the proliferation of B16 melanoma cells, but also promotes the activity of cellular tyrosinase and the synthesis of melanin, which contribute to its whitening mechanism. It can be expected to be used for whitening products.
