摘要
目的优选芪元颗粒中总黄酮的提取和纯化工艺。方法以出膏率和总黄酮得率作为考察指标,采用L_9(3~4)正交试验确定最佳的提取工艺条件;并以pH值、时间、温度、上样量、洗脱溶剂种类和体积为影响因素,通过比较聚酰胺对芪元颗粒中总黄酮的吸附率与解吸率,确定最佳纯化工艺条件。结果最佳的提取工艺:用6倍量70%的乙醇提取2次,每次0.5 h;最佳的纯化工艺:上样量为8 mL(浓度6.28 mg·m L^(-1)),在pH=6,温度为20℃的条件下,控制流速为1 mL·min^(-1),吸附1.5 h,之后用90%的乙醇100 mL进行洗脱。结论所得提取和纯化工艺简单可行,效果良好。
Objective To optimize the extraction and purification process for the total flavonoids from Qi Yuan granule.Methods With yield of extract and total flavonoids as inspection indexes,orthogonal test was adopted to optimize the extraction condition. And with p H value,time,temperature,sample volume,type and volume of elution solvent as the influence factors,the optimum purification condition was screened through the comparison of absorption rate and desorption rate of polyamide to the total flavonoids of Qi Yuan granule. Results The optimum extraction process was extracting 2 times with 6-fold 70 % ethanol, 0.5 hour for each time, and the optimal purification process was achieved by 8 m L of sample volume(concentration being 6.28 mg·m L^(-1)),at pH 6 and 20 ℃,a flow rate of 1 m L·min^(-1),absorption for 1.5 h,and then elution with 100 m L of 90 % ethanol. Conclusion The extraction and purification process is simple and feasible for Qi Yuan granule,and the effect is satisfactory.
