摘要
目的观察结直肠癌组织中miR-150的表达变化,探讨miR-150对结直肠癌细胞增殖的影响。方法选择35例结直肠癌患者,采用实时荧光定量聚合酶链反应(qRT-PCR)检测癌组织和癌旁正常组织中miR-150的表达。将结直肠癌HCT116细胞分为两组,分别采用miR-150模拟物(转染组)和阴性对照(对照组)进行转染,采用CCK-8法检测两组的细胞增殖活性。结果 miR-150在结直肠癌组织中的相对表达量为0.071(0.031,0.129),癌旁正常组织中为0.336(0.168,0.817),癌组织明显低于癌旁正常组织(P<0.05)。与对照组比较,转染组转染48、72、96、120 h时细胞增殖活性显著降低(P均<0.05)。结论结直肠癌组织中miR-150表达下调;miR-150具有抑制结直肠癌细胞增殖的作用,上调miR-150表达可抑制结直肠癌细胞增殖。
Objective To observe the expression changes of miR-150 in colorectal cancer tissues and to investigate the influence of miR-150 on cell proliferation of colorectal cancer. Methods The expression of miR-150 in 35 cases of colorectal cancer tissues and adjacent normal tissues was detected with real-time fluorescent quantitative polymerase chain reaction( qRT-PCR). HCT116 cell line was transfected with miR-150 mimic and negative control respectively,and the cell proliferation activities of the two groups were detected with CCK-8 method. Results The expression of miR-150 in the colorectal cancer tissues and adjacent normal tissues was 0. 071( 0. 031,0. 031) and 0. 336( 0. 168,0. 168),respectively. The expression of miR-150 in the colorectal cancer tissues was significantly lower than that of the adjacent normal tissues( P〈0. 05). Compared with the control group,the activity of HCT116 cell proliferation in the transfection group was significantly decreased at 48 h,72 h,96 h and 120 h( all P〈0. 05). Conclusions The expression of miR-150 in the colorectal cancer tissues is down-regulated. The miR-150 has inhibitory effect on colorectal cancer and the up-regulated expression of miR-150 can inhibit the proliferation of colorectal cancer cells.
