摘要
目的检测鼻咽癌组织和鼻咽癌细胞株中人源含SET结构域蛋白2(SETD2)mRNA的表达,并分析其与EB病毒编码的潜伏膜蛋白1(LMP1)的关系。方法取42例初诊鼻咽癌患者(观察组)和15例慢性鼻咽黏膜炎症患者(对照组)的手术活检组织标本,另取人高分化鼻咽癌细胞株CNE1和稳定转染LMP1基因的CNE1细胞株CNE1-LMP1,采用实时荧光相对定量PCR方法检测观察组、对照组鼻咽组织以及CNE1-LMP1与CNE1中的SETD2 mRNA。结果观察组SETD2 mRNA相对表达量为1.596±0.916,低于对照组的2.194±1.144,且随淋巴结转移分级升高而降低(P均<0.05);CNE1-LMP1中SETD2 mRNA相对表达量为0.676±0.042,低于CNE1的0.870±0.114(P<0.05)。结论 SETD2在鼻咽癌组织中的表达下调,可能与鼻咽癌的发生发展有关,SETD2基因表达下调可能与EB病毒编码的LMP1有关。
Objective To detect the SETD2 mRNA expression in the nasopharyngeal carcinoma( NPC) tissues and NPC cell line,and to explore its relationships with latent membrane protein 1( LMP1) coded by EB virus. Methods Real-time fluorescent relatively quantitative PCR was used to detected SETD2 mRNA expression in 42 nasopharyngeal carcinoma tissues( observation group),15 non-cancerous nasopharyngeal tissues( control group),and high-differentiated nasopharyngeal carcinoma cell lines CNE1 and CNE1-LMP1. Results The SETD2 mRNA expression of the observation group( 1. 596 ± 0. 916) was lower than that of the control group( 2. 194 ± 1. 144)( P〈0. 05),and decreased with the increased lymph node metastasis grading. The SETD2 mRNA expression of CNE1-LMP1( 0. 676 ± 0. 042) was lower than that of CNE1( 0. 870 ± 0. 114)( P〈0. 05). Conclusions The declined expression of SETD2 mRNA indicated that it might be involved in the development and progression of nasopharyngeal carcinoma. The down-regulation of SETD2 gene was related with EBV-LMP1 to some degree.
