摘要
目的:采用核磁共振技术、TTC染色方法检测葛根素预处理对脑缺血急性期的保护作用。方法:48只SD大鼠随机分为6组,每组8只。假手术组和模型对照组给予生理盐水,受试药给予葛根素注射液(21、42、84 mg/kg),阳性对照组给予灯盏细辛注射液(18 mg/kg)。尾静脉注射给药7天预防治疗后进行血栓栓塞性脑缺血手术。术后6 h行神经功能评分,采用核磁共振技术进行大鼠脑SE-T2WI以及弥散加权成像(DWI)序列扫描,测量脑病变范围;术后24 h评分后取脑进行TTC染色,测量脑梗死范围;取血清检测SOD、MDA。结果:葛根素42mg/kg预处理组大鼠缺血6 h和24 h行为障碍分别改善17.8%、33.3%;6 h脑病变范围和24 h梗死范围缩小29.4%、31.6%,血清MDA含量降低16.0%,SOD活力升高21.1%;葛根素84mg/kg较42mg/kg效果稍好,葛根素21mg/kg未见明显效果。结论:葛根素预处理能改善大鼠脑缺血急性期受损神经功能,缩小脑病变范围,具脑保护作用。
Objective: To observe the effects of Puerarin preconditioning on the thromboembolic cerebral ischemia by Magnetic Resonance Imaging ( Mill), Trc staining and pathological methods. Methods: Forty-eight SD rats were randomly divided into 4 groups ( n = 8 ). Sham group and the model group were given saline, the reagent and positive control group were given puerarin injection (21,42,84mg/kg) and breviscap- ine injection(4 ml/kg) for 7 days. Then the left middle cerebral artery of all rats was occluded by autologous blood clots. At 6h after MCAO, neurological deficit score and MRI including SE-T2WI and DWI sequence scanning were measured. At 24 h after MCAO, the brain was cut for TYC staining and the serum was obtained to measure the oxidative stress after neurological deficit score. Results: At 6 h and 24 h, Puera- rin d2mg/kg preconditioning improved behavior disorder by 17.8% ( P 〉 0.05 ) and 53.3% ( P 〈 0.05 ) ; decreased brainlesion area and in- farction scope by 29.4% ( P 〈 0.05 ) and 31.6% ( P 〈 0.05 ) respectively, and reduced serum MDA content by 16.0% ( P 〈 0.05 ), in- creased SOD activity by 21. 1% (P 〈 0.05 ), Puerarin 84mg/kg had better effect than 42mg/kg slightly, puerarin 21mg/kg showed no effect. Conclusion: Puerarin pretreatment can improve the damaged neural function, decrease extent of disease, relieve the oxidative stress, with the cerebral protective effects for cerebral ischemia of rats.
