摘要
目的:研究麻黄及其与桂枝配伍前后麻黄生物碱在大鼠体内组织分布变化的特征,并对二者的分布情况进行对比研究。方法:以金刚烷胺为内标,采用UPLC-MS/MS法测定大鼠灌胃给予麻黄单煎液及麻黄-桂枝药对水煎液后三个时间点(0.25、1.5、3h)组织样品中5种麻黄生物碱的浓度。结果:与麻黄组相比,给药后0.25h,不同程度的降低去甲基麻黄碱、麻黄碱和伪麻黄碱在各组织的含量,增加了去甲基伪麻黄碱和甲基麻黄碱在各组织的含量;在分布的平衡期(1.5h),麻黄桂枝组均能显著提高麻黄类生物碱在肺组织中的含量,提高去甲基麻黄碱、去甲基伪麻黄碱和甲基麻黄碱在肾组织中的含量;在给药后3h,麻黄桂枝组均能显著提高麻黄类生物碱在脑组织中的含量,显著提高去甲基麻黄碱、麻黄碱和伪麻黄碱在肾组织中的含量,显著降低去甲基麻黄碱、麻黄碱、伪麻黄碱和甲基麻黄碱在心组织的含量。结论:桂枝增加了麻黄类生物碱在大鼠脑组织中的分布,增加去甲基麻黄碱、麻黄碱和伪麻黄碱在肾中的分布;降低了去甲基麻黄碱、麻黄碱、伪麻黄碱和甲基麻黄碱在心组织的分布。
Objective: To study the tissue distribution features of ephedra alkaloids in rat before and after the compatibility of Mahuang and Guizhi, and compare the difference of distribution of ephedra alkaloids under the two administration models. Methods: With amantadine as internal standard, UPLC-MS/MS method was used to determine the concentration of five kinds of ephedra alkaloids in tissue samples in three different time points after given decoction of Mahuang and Mahuang-Guizhi herb pair. Results: Compare with Mahuang group, at 0.25h after adminis- tration, significantly lower the concentration of norephedrine, ephedrine and pseudoephedrine in tissue, and increase the concentration of norpseudoephedrine and methylephedrine in tissue; In the equilibrium phase of tissue distribution (1.5h), Mahuang-Guizhi group can signif- icantly improve the concentration of ephedra alkaloids in lung, and also can improve the concentration of norephedrine, norpseudoephedrine and methylephedrine in kidney; At 3h after administration, Mahuang-Guizhi group can significantly improve the concentration of ephedra al- kaloids in brain, also can improve the concentration of norephedrine, ephedrine and pseudoephedrine in kidney, and can significantly reduce the concentration of norephedrine, ephedrine, pseudoephedrine and methylephedrine in heart. Conclusion : There have different degree im- pact of Guizhi on ephedra alkaloids of tissue distribution in rats of Mahuang-Guizhi herb pair.
