摘要
目的:探讨制首乌提取物对兔动脉粥样硬化模型中ABCA1表达的影响。方法:雄性新西兰大白兔36只,随机分为正常组、模型组、制首乌提取物15mg/kg组、制首乌提取物7.5mg/kg组、制首乌提取物3.75mg/kg组和血脂康400mg/kg组6个组,使用免疫损伤法和高脂高胆固醇饲料喂养新西兰大白兔12周,建立兔动脉粥样硬化模型。给药4周后,耳缘静脉采血检测血清TG、TC、LDL和HDL水平。处死动物,取主动脉弓下约2cm处胸主动脉进行HE染色后于光镜下观察,并采用RT-PCR法测定胸主动脉中ABCA1mRNA的表达。结果:病理学切片检查显示,制首乌提取物能够延缓动脉粥样硬化病变进程。RT-PCR结果显示,与模型组相比各剂量的制首乌提取物能不同程度上调胸主动脉中ABCA1 mRNA的表达。结论:制首乌提取物具有降血脂及抗动脉粥样硬化作用,该抗动脉粥样硬化作用机制可能与其对ABCA1的上调有关。
Objective: To discuss the influence extract of Radix Polygoni muhiflori Preparata(RPMP) has on ABCA1 expression in rabbit arterio- sclerosis models. Method : 36 male New Zealand rabbits were assigned to 6 groups : normal group, model group, high dose group with extract of RPMP 15mg/kg, medium dose group with extract of RPMP 7.5mg/kg, low dose group with extract of RPMP 3.75mg/kg and Xuezhikang 400mg/kg group. The rabbits were fed with immune injury method and high fat high cholesterol fodder for 12 weeks to build rabbit arterioscle- rosis models. After 4 weeks, blood samples were taken from ear veins to test serum TG, TC, LDL and HDL. All the animals were killed and samples of thoracic aorta were taken 2 cm below the arcus aortae. The samples were observed in light microscope after HE staining and AB- CA1 mRNA expression in thoracic aorta were measured with RT-PCR. Results: Pathology sections showed that extract of RPMP could slow down the progress of AS. RT-PCR results showed that extract of RPMP up-regulated the expression of ABCA1 mRNA in thoracic aorta compa- ring with model group. Conclusion: Extract of RPMP could reduce blood fat and had the effect of anti-atheroselerosis and the mechanism may be related with the up-regulation of ABCA1.
