癫痫患者脑脊液和血清中miRNAs含量检测及其对相关细胞因子的调控作用 被引量:16

摘要 目的:研究癫痫患者脑脊液和血清中miRNAs的含量及其对相关细胞因子的调控作用。方法:采集癫痫组和非癫痫组患者的血清和脑脊液,检测miR-146a、miR-221、miR-222、miR-21、miR-34a以及炎性因子、凋亡分子的含量。结果:癫痫组脑脊液中miR-146a、miR-221、miR-222、miR-21的含量显著低于非癫痫组,miR-34a的含量高于非癫痫组;血清中miR-146a、miR-221、miR-222、miR-21的含量显著高于非癫痫组,miR-34a的含量低于非癫痫组;癫痫组脑脊液中TNF-α、IL-2、IL-6、ICAM-1的含量高于非癫痫组且与miR-146a、miR-221、miR-222的含量呈负相关;癫痫组脑脊液中Bax、Bim、Caspase-3、Caspase-9的含量高于非癫痫组且与miR-21呈负相关,Bcl-2的含量低于非癫痫组且与miR-34a呈负相关。结论:癫痫患者脑脊液中miR-146a、miR-221、miR-222、miR-21含量降低而miR-34a含量升高,miRNAs能够调节炎性因子、凋亡分子的表达并参与神经元功能的改变。 Objective: To detect the contents of miRNAs in cerebrospinal fluid and serum of patients with epilepsy and their regulating effects on related cytokines. Methods: Serum and cerebrospinal fluid of epilepsy group and non-epilepsy group were collected to detect the contents of miR-146a, miR-221, miR-222, miR-21 and miR-34a as well as inflammatory factors and apoptosis molecules. Results: miR-146a, miR-221, miR-222 and miR-21 contents in cerebrospinal fluid of epilepsy group were significantly lower than those of non-epilepsy group, but miR-34a content was higher than that of non-epilepsy group; miR-146a, miR-221, miR-222 and miR-21 contents in serum were significantly higher than those of non-epilepsy group, and miR-34a content was lower than that of non-epilepsy group; TNF-α, IL-2, IL-6 and ICAM-1 contents in cerebrospinal fluid of epilepsy group were higher than those of non-epilepsy group and negatively correlated with miR-146a, miR-221 and miR-222 contents; Bax, Bim, Caspase-3 and Caspase-9 contents in cerebrospinal fluid of epilepsy group were higher than those of non- epilepsy group and were negatively correlated with miR-21. Bcl-2 content was lower than that of non-epilepsy group and negatively correlated with miR-34a. Conclusion: miR-146a, miR-221, miR-222 and miR-21 contents decrease while miR-34a content increases in cerebrospinal fluid of patients with epilepsy, and miRNAs can regulate the expression of inflammatory factors and apoptosis molecules and it is involved in the changes of neuron function.
