摘要
为了解新疆玛纳斯小海子水库周边典型湿地的放线菌的群落结构及遗传的多样性,本研究采用变性梯度凝胶电泳(denaturing gradient gelelectrophoresis,DGGE)技术对玛纳斯湿地土壤中放线菌群落的16S r DNA V3区进行电泳分离,并用Quantity one软件对图谱进行分析,对主要条带进行回收测序及序列比对并构建系统发育树。DGGE图谱显示,植物丰富度不同的区域中土壤放线菌的多样性指数(H)、丰富度(S)和均匀度(EH)有显著的差异(P<0.5),放线菌的多样性指数、丰度均为:沼泽土>草甸土>盐碱土。特异性条带回收测序结果表明,链霉菌属属于该区域放线菌中的最优菌群。
To understand the genetic diversity and community structure of actinomyeetes in the typical wetland around the Manas Xiaohaizi reservoir of xinjiang,denaturing gradient gel electrophoresis (DGGE) technique was applied to separate the 16S r DNA V3 region of soil actinomycetes community the Manas wetland in this study.We then use the Quantity one software to analyze the map patterns.The main bands of the DGGE were recovered,sequenced,and the nueleotide sequences were aligned.A phylogentic tree was constructed based on the alignment resuhs.DGGE profiles showed that the actinomycetes diversity index (//),richness (S) and evenness (EH) in the different areas of plant species richness have a significant difference (P〈0.5). Actinomycetes diversity index (/4),abundance (S) are the marsh soil〉 meadow soil〉 saline soil.The sequenced results of the specific bands recovered show that the Streptomyces is the optimal flora the actinomyeetes in the region.By DGGE more objectively reflect the Manas Xiaohaizi around the reservoir typical wetland actinomycetes bacteria community structure diversity.
