清开灵软胶囊制剂前体体外抗H1N1、H5N1和H7N9流感病毒作用

Effect of Qingkailing(清开灵)Soft Capsule Precursor on H1N1,H5N1 and H7N9 Influenza Virus in Vitro

目的观察清开灵软胶囊制剂前体体外抗H1N1、H5N1、H7N9流感病毒的作用。方法通过测定细胞培养液上清血凝滴度(HA)和半数组织培养感染剂量(TCID_(50)),评价清开灵软胶囊制剂前体和利巴韦林注射液不同给药方式(灭活、治疗、中和、预防)对H1N1、H5N1和H7N9流感病毒的抑制作用,并且通过MTT法测定清开灵软胶囊制剂前体和利巴韦林注射液不同给药方式体外抗H1N1流感病毒作用。结果清开灵软胶囊制剂前体的最大无毒浓度为0.391 mg/ml,利巴韦林注射液的最大无毒浓度为3.907μg/ml。清开灵软胶囊制剂前体以预防方式给药可以明显抑制流感病毒增殖,能显著降低H1N1、H5N1和H7N9流感病毒的HA和TCID_(50),以治疗方式给药能明显降低H1N1流感病毒的HA和TCID_(50)(P<0.05或P<0.01)。清开灵软胶囊制剂前体和利巴韦林注射液抗H1N1流感病毒的半数有效浓度分别为0.25 mg/ml、2.19μg/ml,治疗指数分别为5.32、95.89。结论清开灵软胶囊制剂前体对H1N1、H5N1、H7N9流感病毒具有明显的预防作用,且对H1N1流感病毒具有良好的治疗作用。

Objective To observe the effect of Qingkailing soft capsule precursor against H1N1,H5N1,H7N9 influenza virus in vitro.Methods Through determination of cell culture supernatant of hemagglutination titer（HA）and median tissue culture infective dose（TCID50）,inhibition of influenza virus H1N1,H5N1,and H7N9 would be evaluated of Qingkailing soft capsule precursor and Ribavirin injection with different administration way（inactivated,treatment,neutralization and prevention）,the antiviral effect of Qingkailing soft capsule precursor and Ribavirin injection against the H1N1 influenza virus in different ways of administration in vitro will be determined by MTT method.Results The maximum non-toxic concentration of Qingkailing soft capsule precursor is 0.391 mg/ml and 3.907 μg /ml for Ribavirin injection.Qingkailing soft capsule precursor can significantly inhibit the proliferation of influenza virus with drug prevention way,it can significantly reduce HA and TCID50 of H1N1,H5N1 and H7N9 influenza virus,and it can significantly reduce HA and TCID50 of H1N1 influenza virus with drug treatment way（P〈0.05 or P〈0.01）.The median effective concentration of Qingkailing soft capsule precursor and Ribavirin injection against H1N1 influenza virus is 0.25 mg/ml,2.19 g/ml respectively.The therapeutic index is 5.32,95.89 respectively.Conclusion Qingkailing soft capsule precursor has significant preventive effect against influenza virus of H1N1,H5N1 and H7N9,and it has a good therapeutic effect for H1N1 influenza virus.
