摘要 目的 确立HPLC-DAD法同时测定龙胆泻肝丸中3种活性成分的含量. 方法 PLATSILTM ODS柱(250 mm x 4. 6 mm,5 μm);流动相为甲醇( A)-0. 1%磷酸,梯度洗脱,流速为1 mL/min,检测波长分别为280 nm(龙胆苦苷、黄芩苷)和240 nm(栀子苷);柱温30 ℃. 结果 龙胆苦苷、栀子苷和黄芩苷线性范围分别为0. 112-2. 016μg(r=1. 000 0)、0. 007 6-0. 136 8μg(r=0. 999 8)、0. 294-5. 292μg(r=0. 999 7). 范围内与峰面积呈良好的线性关系,平均回收率分别为100 . 5%、99 . 9%和104 . 5%. 结论 该方法准确、简便、具有良好的重现性和稳定性,适用于龙胆泻肝丸这种药品的质量控制,提高药品的质量. Objective To establish a method for simultaneous determination of gentiopicroside,berberine and baica-lin in Longdan Xiegan pills. Methods HPLC-DAD method was employed to determine three active ingredients in Longdan Xiegan pills simultaneously. Using PLATISILTM ODS (250 mmx 4. 6 mm,5 μm) column with methanol (A)-0. 1% phosphoric acid(B) gradient:0-5 min 70%(B),5-45 min 70% -20%,a flow rate of 1 mL/min,detec-tion wavelength was 280 nm for gentiopicroside and baicalin,while 240 nm for geniposide,and the column temperature was 30 ℃. Results The results showed that the linear range of gentiopicroside, geniposide and baicalin was 0. 112-2. 016 μg(r=1. 000 0),0. 007 6-0. 136 8 μg(r=0. 999 8),0. 294-5. 292 μg(r=0. 999 7) respectively and the peak area with the range appeared a good linear relationship,The average recoveries (n=6) were 100. 5%, 99. 9% and 104. 5%. Conclusion The method is accurate,simple,reproducibility and stability,which avaliable for quality control of Longdan Xiegan pills.
