耳聋左慈丸对老年性耳聋模型小鼠血清SOD、MDA、PKC的影响 被引量:10

摘要 目的研究耳聋左慈丸对老年性耳聋模型小鼠血清SOD、MDA、PKC含量的影响,探讨治疗老年性耳聋的机制。方法将100只健康雄性SPF级昆明小鼠随机分为2组,对照组(CG)15只和造模组85只,造模组采用腹腔注射D-半乳糖(150 mg/kg.d),对照组腹腔注射等剂量0.9%生理盐水,每日1次,连续2个月。造模成功后,将75只小鼠随机分为5组,模型组(MG)、VitC组(VCG)、中药低剂量组(LDG)、中药中剂量组(MDG)、中药高剂量组(HDG),每组15只。VCG灌胃Vit C注射液(150 mg/kg.d),中药治疗组灌胃不同浓度耳聋左慈丸溶液,分别是LDG(0.3 g/ml)、MDG(0.6 g/ml)、HDG(1.2g/m)l,CG和MG灌胃等剂量的0.9%NaC l,每日1次,连续14天。采用听性脑干反应(ABR)检测听力水平,用比色法测定血清的SOD活性、MDA含量的变化,用双抗体夹心-酶联免疫吸附(ABC-ELISA)法检测血清中PKC的含量。结果与对照组相比,模型组小鼠的一般情况均减退,且各频率ABR值显著增高(P<0.01或P<0.05),血清SOD活性降低、血清MDA含量增高(P<0.01)、血清PKC的含量升高(P<0.01)。与模型组相比,中药组小鼠的一般情况均得到改善,且各频率ABR值降低(P<0.01),依次是HDG<MDG<LDG,血清SOD活性升高,依次是HDG>MDG>LDG,血清MDA含量逐渐降低,依次是LDG>MDG>HDG,,减轻了氧自由基引起的病理损伤;VCG小鼠血清的SOD活性、MDA含量与LDG相近。中药各组显著降低血清PKC的含量(P<0.01)。结论耳聋左慈丸可以减轻老年性耳聋模型小鼠的病理损害,对氧自由基的损伤有一定的保护作用;降低PKC浓度,从而对c AMP/PKC信号通路起调节作用。 Objective To research the effect of the Erlong Zuoci pill on SOD, MDA and PKC in serum of mice with presbycusis mod- el and the related mechanism. Methods One hundred healthy male SPF Kunming mice were randomly divided into 2 groups: control group (CG) had fifteen mice, and molding group had eighty-five mice. The mice in CG were treated with 0.9% NaC1 and the other mice were treated with ip D-galactose (150mg/kg.d) , once a day, with 2 months. After duplicating the mold, the seventy-five mice were randomly divided into 5 groups: model group (MG) , VitC group (VCG) , Chinese medicine low dose group (LDG) , Chinese medicine mid-dose group (MDG) , Chinese medicine high dose group (HDG) , with 15 mice ineach group. The mice in VCG were treated withVitC injection (150mg/kg.d) , the Chinese medicine group were treated with different density Erlong Zuoci pills, included LDG (0.3g/ml) , MDG (0.6g/ml) , HDG (1.2g/ml) once a day, with 14 days. We checked its listening ability by auditory brainstem response (ABR) , and used colofimetric method to determine the changes of the serum SOD activity and MDA content and the PKC in rats serum was detected by double antibody clip ELASA method. Results Compared with CG, the general condition of mice was subsided, the value of ABR in MG was increased markedly (P〈0.01 or P〈0.05) , the serum SOD activity decreased, and MDA content increased, PKC content increased (P〈0.01) . Compared with MG, the general condition of mice was improved, the val- ue of ABR in the Chinese medicine group was decreased (P〈0.01 or P 〈0.05) , HDG 〈 MDG〈 LDG, the serum SOD activity was decreased, HDG 〉MDG 〉 LDG, and MDA contentlincreased (P〈0.01) , LDG 〉 MDG 〉 HDG, and the SOD, MDA of LDG and VCG is similar. The PKC content of traditional medicine group were significantly decreased (P〈0.01) . Conclusion The Erlong Zuoci pill can reduce the pathologic lesions of mice with presbycusis, protect from the damage caused by oxygen free radicals. The Erlong Zuoci pill could decrease PKC content and thus !aalv a regulatory role in the function of cAMP/PKC.
