摘要
探讨miR-1对牛骨骼肌卫星细胞增殖的影响,为改善牛肉品质提供重要的理论基础。首先构建miR-1的过表达载体并进行牛骨骼肌卫星细胞的转染,然后采用Ed U法检测发现miR-1抑制牛骨骼肌卫星细胞的增殖。双荧光素酶报告表明PAX3为miR-1的靶基因。最后检测出miR-1过表达后PAX3在mRNA和蛋白表达水平均下降。试验结果提示:miR-1能够通过抑制PAX3的表达而抑制牛骨骼肌卫星细胞的增殖。
In order to explore effects of miR- 1 on bovine skeletal muscle cell proliferation,to help improving the quality of beef. We transfect miR- 1 into bovine skeletal muscle satellite cells and test the conditions of cell proliferation by Ed U kit. Results showed that PAX3 was the target genes of miR- 1. MiR- 1 inhibited the expression of PAX3 at the level of mRNA and protein. The results suggested that miR- 1 inhibited muscle cell proliferation via inhibiting the expression of PAX3.
